摘 要:目的 :观察新风胶囊对佐剂性关节炎的治疗作用及对细胞因子系列 (IL 1、IL 4、IL 10及TNFα)的影响。方法 :将 5 0只大鼠随机分为正常对照组、模型对照组、新风胶囊组、雷公藤多苷片(TPT)组和甲氨喋呤 (MTX)治疗组 ,每组 10只 ,制备完全佐剂并分别向除正常对照组以外的其余动物右后足跖皮内注射 0 .0 5ml致炎。观察各组血清中各细胞因子含量的变化。结果 :与治疗前比较 ,新风胶囊治疗组、TPT治疗组及MTX治疗组治疗后大鼠的关节炎指数 (AI)显著降低 (P <0 .0 5或P <0 .0 1)。新风胶囊治疗组给药后体质量增加值与正常对照组比较差异无显著性 (P >0 .0 5 ) ;TPT治疗组及MTX治疗组给药前后体质量增加值显著低于正常对照组和新风胶囊治疗组(P <0 .0 1)。与模型对照组相比 ,新风胶囊治疗组的IL 1、TNFα显著降低 ,IL 4、IL 10显著升高 (P <0 .0 1) ,并与其余两治疗组无显著性差异 (P >0 .0 5 )。结论 :新风胶囊与MTX、TPT一样能降低佐剂性关节炎模型大鼠的AI ,而在增加大鼠体质量方面优于TPT和MTX ;新风胶囊可能通过下调致炎因子 (IL 1、TNFα)、上调抗炎因子 (IL 4、IL 10 )、抑制致炎效应、增强抗炎效应 ,从而达到降低AI、消除肿胀的治疗目的。Objective:To observe the effects of Xinfeng capsule(XFC) on the levels of interleukin 1(IL 1),tumor necrosis factor α (TNFα), interleukin 4 (IL 4), interleukin 10 (IL 10) of adjuvant arthritis rats. Method: 50 rats were averagely divided into normal control group, model control group, XFC treated group, tripterygium wilfordii polycoride tablet (TPT) treated group, methotrexate (MTX) treated group. Except for the rats of normal control group, the others were intracutaneously injected with 0.05 ml of Freung′s complete adjuvant in the right hindlimb. The serum cytokine levels of each rat were assayed. Result:The arthritis index (AI) of XFC, TPT,MTX treated group was markedly decreased after treatment (P<0.05 or P<0.01). The increment of rat body weight (BW) in XFC treated group wasn′t significantly different from that of normal control group (P>0.05). The increment of rat BW of TPT, MTX treated group was significantly lower than that of XFC treated group, normal control group (P<0.01). The IL 1 level of XFC treated group was ob viously decreased in comparison with that of model control group, and IL 4,IL 10 level significantly increased (P<0.01). There was no significant difference between the IL 1, TNFα, IL 4,and IL 10 levels of XFC treated group and the other two treatment group (P>0.05). Conclusion: The effect of XFC on the decrement of AI is the same as that of TPT, MTX in adjuvant arthritis rats, while effect of XFC on the increment of BW is superior to that of TPT, MTX. XFC decreases AI, eliminates swelling by down regulating the levels of IL 1, TNFα, up regulating the levels of IL 4, IL 10, restraining the inflammatory reaction.
