摘 要:目的 探讨 B1 9病毒中国株的基因变异 .方法 采用长片段聚合酶链反应技术 ,从中国再障患儿血清中扩增HPVB1 9- XA2株的 VP1独特区基因和 VP1 /VP2基因片段 ,进行核苷酸序列分析 .结果 B1 9- XA2株的 VP1独特区和VP1 /VP2连接区基因大部分区域核苷酸序列被测定 ,其与国外 Au株基因核苷酸序列比较 ,有 4个核苷酸突变 ,并引起编码的 2个氨基酸改变 .结论 我国 B1 9病毒 VP1基因独特区和 VP1 /VP2连接区基因序列与AIM To explore the gene structure and mutation of HPVB19 (human parvovirus B19) isolates obtained from Chinese patients with aplastic crisis. METHODS Specific genes of B19 XA2 strain from an aplastic crisis of Chinese child were amplified by PCR and then the amplification products were sequenced directly. RESULTS B19 XA2 VP1 gene of unique region and VP1/VP2 region were obtained by long distance PCR and nest PCR, and the data of nucleotide sequence analysis was compared with sequences of isolated B19 Au, the B19 XA2 had 4 bases changes and 2 sequencesof their coding amino acid was changed also. CONCLUSION Genetic diversity exists in the nucleotide sequence of a VP1 specific gene and VP1/VP2 gene of HPV B19 isolated from the serum of Chinese child.
