摘  要:目的探讨癌/睾丸抗原TEX14与乳腺癌转移及患者预后的关系。方法利用Oncomine网站对目前已知的所有癌/睾丸抗原(CTAs)、促进乳腺癌转移基因AKT2、CD44、EIF4E、ERBB2及抑制乳腺癌转移基因BRMS1、CD82、KISS1、TIMP1在侵袭性和非侵袭性乳腺导管癌中m RNA表达情况进行筛查和分析。利用免疫组化方法检测乳腺癌组织标本的癌细胞TEX14蛋白表达情况。利用TCGA数据库1 102例乳腺癌样本mRNA高通量测序数据,分别对TEX14与多个乳腺癌转移相关基因mRNA表达量进行相关性分析。采用Kaplan-Meier生存曲线和Logrank检验法分析TEX14的表达与乳腺癌患者预后的关系。结果与非侵袭性乳腺导管癌相比,在侵袭性乳腺导管癌中,86个CTAs mRNA表达量升高,其中TEX14升高倍数最大为3.344倍(P<0.05);促进乳腺癌转移基因AKT2、CD44、EIF4E、ERBB2 mRNA表达量升高,且与TEX14的表达呈正相关(P<0.05);抑制乳腺癌转移基因BRMS1、CD82、KISS1、TIMP1 mRNA表达量降低,且与TEX14的表达呈负相关(P<0.05)。进一步利用免疫组化实验证明,与无转移组相比,有转移组乳腺癌细胞TEX14阳性细胞率升高1.5倍(P<0.05)。TEX14高表达组乳腺癌患者总生存期较低表达组明显缩短(P<0.05)。结论癌/睾丸抗原TEX14在侵袭转移性乳腺癌中高表达,与患者的不良预后相关,为进一步研究TEX14可促进乳腺癌转移提供了实验依据。Objective The aim of this study was to analysis the relationship between cancer/testis antigen TEX14 and breast cancer metastasis and prognosis. Methods The reports of mRNA expression of all currently known cancer/testis antigens(CTAs) and promoting breast cancer metastasis genes AKT2,CD44,EIF4 E,ERBB2,and inhibiting breast cancer metastasis genes BRMS1,CD82,KISS1,TIMP1 in non-invasive and invasive ductal breast carcinomas were obtained from the Oncomine website. Immunohistochemistry was used to detect the protein expression of TEX14 in breast cancer cells. The data of gene expression profiles of 1 102 breast cancer samples obtained from TCGA database was used for the correlation analysis of the mRNA expression levels of breast cancer metastasis related genes and TEX14. Kaplan-Meier survival curve and Log-rank test were used to analyze the correlation between TEX14 expression and the prognosis of breast cancer patients. Results Compared with non-invasive ductal breast carcinoma,the m RNA expression levels of 86 CTAs increased in invasive ductal breast carcinoma and the maximal increase of TEX14 was 3.344 times(P<0.05). The m RNA expression levels of promoting breast cancer metastasis genes,AKT2,CD44,EIF4 E and ERBB2,were increased and that were positively correlated with TEX14(P<0.05). The mRNA expression levels of inhibiting breast cancer metastasis genes,BRMS1, CD82, KISS1 and TIMP1, were decreased and that were negatively correlated with TEX14(P<0.05).Immunohistochemistry was further used to show that the rate of TEX14-positive cells in metastatic group was 1.5 times higher than that in non-metastasis group(P<0.05). The overall survival time of TEX14 high-expressing breast cancer patients was significantly shorter than that of TEX14 low-expressing group(P<0.05). Conclusion Cancer/testis antigen TEX14 was highly expressed in invasive breast cancer and associated with a poor prognosis in patients. It provided an experimental basis for further study of TEX14 which could promote breast cancer metastasis.
