摘  要:目的探讨熟地黄多糖耙向肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)/信号转导及转录活化因子3(activator of transcription 3,STAT3)通路抑制鼻咽癌增殖转移的机制。方法以鼻咽癌CEN1细胞为研究对象,分别以50μg/m L(A组)、100μg/m L(B组)、400μg/m L(C组)和800μg/m L(D组)浓度的熟地黄多糖溶液处理,MTT实验绘制处理前、处理24、48和72 h的CEN1细胞增殖曲线,同期采用RT-PCR测定各组TNF-α、STAT3和Janus蛋白酪氨酸激酶(Janus protein tyrosine kinase,JAK)的mRNA转录水平,并采用Transwell小室体外迁移实验观察处理前和处理72 h CEN1细胞转移能力的变化。结果不同浓度的熟地黄多糖处理,对鼻咽癌CEN1细胞的增殖具有抑制作用,且呈现明显的剂量依赖性和时间依赖性。不同浓度的熟地黄多糖处理,对鼻咽癌CEN1细胞的转移具有抑制作用,且呈现明显的剂量依赖性和时间依赖性。不同浓度的熟地黄多糖处理鼻咽癌CEN1细胞,B组、C组和D组TNF-α、STAT3和JAK的mRNA相对表达量随着时间的延长而降低,且其对TNF-α、STAT3和JAK的mRNA转录水平调控效果具有明显的剂量依赖性。结论熟地黄多糖抑制鼻咽癌增殖转移具有一定的时间和剂量依赖性,而下调TNF-α、STAT3和JAK可能为其抑制鼻咽癌增殖转移的机制。Objective To investigate the roles of tumor necrosis factor α(TNF-α)/signal transducer and activator of transcription 3(STAT3) pathway in radix rehmanniae polysaccharide inhibiting nasopharyngeal carcinoma(NPC)cell proliferation and metastasis.Methods NPC cell line CEN1 was taken as the research object,and treated with rehmannia polysaccharide solution of 50 g/m L(Group A),100 g/m L(Group B),400 g/m L(Group C) and 800 g/m L(Group D).Before and after treatment for 24 h,48 h and 72 h,CEN1 cells proliferation was determined by MTT.RT-PCR was applied in analyze the mRNA levels of TNF-α,STAT3 and Janus protein tyrosine kinase(JAK).Trans-well cell migration was applied for metastasis of CEN1 cells before and 72 h after treatment.Results Radix rehmanniae polysaccharide at different concentrations inhibited the CEN1 cell proliferation in dose-dependent and time-dependent manners.Radix rehmanniae polysaccharide at different concentrations significantly inhibited the metastasis of CEN1 cells in dose-dependence and time-dependence manners.In Group B,C and D,radix rehmanniae polysaccharide inhibited TNF-α,STAT3 and JAK mRNA expression in time-dependent and dose-dependent manners.Conclusion Radix rehmanniae polysaccharide inhibits the proliferation and metastasis of NPC in time-and dose-dependent manners,which may be via the down-regulation of TNF-α,STAT3 and JAK.
