摘 要:目的:建立RP-HPLC法测定广东道地药材巴戟天中游离氨基酸和结合氨基酸含量的方法,为其资源开发及药效物质研究提供参考。方法:采用超声辅助提取和微波消解法分别制得游离氨基酸和水解氨基酸溶液,以异硫氰酸苯酯(PITC)为柱前衍生试剂,采用反相高效液相色谱法(RP-HPLC)测定氨基酸含量。结果:17种氨基酸在试验浓度范围内,线性关系良好,相关系数r≥0.9995,游离氨基酸加样回收率为96.80%~101.63%,RSD%为1.61%~3.71%,水解氨基酸加样回收率为97.97%~104.92%,RSD%为0.62%~3.21%。结论:该测定方法简便快捷、精密度高、重复性好,可用于巴戟天中游离氨基酸和结合氨基酸含量测定。Objective:To establish an RP-HPLC method for the content determination of free amino acids and bound amino acids in Bajitian(morinda officinalis),a geo-authentic crude drug,and provide reference for its resource development and medicinal material foundation.Methods:The ultrasonic assisted extraction and microwave digestion were applied to prepare free amino acids and amino acid hydrolysis solution,with phenyl isothiocyanate(PITC) as a pre column derivatization reagent.Then the content of amino acids were detected by reversed-phase high performance liquid chromatography(RP-HPLC).Result:The assay displayed a good linearity in the range of test concentration.The correlation coefficient r≥0.9995.The recovery rates of free amino acids were between 96.80% and 101.63%,and the RSD% values were between 1.61% and 3.71%.The recovery rates in hydrolyzed amino acids were between 97.97% and 104.92%,the RSD% values were between 0.62% and3.21%.Conclution:The method is simple,rapid,accurate and reproducible.It can be used for the determination of free and bound amino acids in Bajitian(morinda officinalis).
