摘 要:目的:运用基因芯片技术研究温阳消饮法对胸腔积液模型大鼠肾髓质差异基因表达的影响。方法:40只清洁级Wistar大鼠随机分为温阳消饮组、去桂消饮组、模型组、空白组,每组10只。除空白组外,其余3组大鼠以1%λ-角叉菜胶制备胸腔积液模型,造模成功后2h,温阳消饮组、去桂消饮组予以相应药物灌胃,日1次,连续5d。灌胃结束后处死大鼠。每组取3只大鼠的肾髓质进行基因表达谱检测,筛选出差异表达基因,进行Pathway分析。结果:(1)温阳消饮组与模型组比较、模型组与空白组比较,其共同表达通路有3条:神经活性配体受体相互作用通路、系统性红斑狼疮通路、肾素血管紧张素系统通路;共同表达基因有7个:LOC317471、Olr462、Mcpt8I3、Olr718、Mcpt8I2、Cd80、Ttn。(2)去桂消饮组与模型组比较、模型组与空白组比较,其共同表达通路有1条:神经活性配体受体相互作用通路;共同表达基因有3个:Cep295nl、Mcpt8I3、Fam64a。结论:(1)温阳消饮法对胸腔积液模型大鼠肾髓质基因的调控是通过多个通路及基因相互作用的结果;(2)该法可能是通过上调Mcpt8I3基因调控神经活性配体受体相互作用通路,影响其炎症反应的;(3)温阳消饮组与去桂消饮组在肾髓质基因的Pathway分析中存在差异,温阳消饮方可能是通过上调Mcpt8I3基因调控肾素血管紧张素系统通路而发挥利尿消饮作用的;(4)本研究中未发现该法对cAMP-PKA/PKC及VWP通路有影响。Objective:To investigate the effect of warming yang for resolving fluid retention therapy on differential gene expression in rats with pleural effusion.Methods:Forty Wistar rats were randomly divided into 4groups:Wenyang Xiaoyin group (W group),Qugui Xiaoyin group (Q group),model group (M group),normal group (N group).Except for the control group,other groups were all established the pleural effusion model by a single intrapleural injection of 1% λ-Carrageenan.Two hours after successful modeling,W group and Q group were given gastric lavage with corresponding drugs,1 time a day for 5 consecutive days.After the end of gavage,rats were sacrificed.Three rats in each group were selected for gene expression profiling,and the differentially expressed genes were screened out for pathway analysis.Results:①Comparing group W with group M,group M with group K,there were three common expression pathways:neuroactive ligand-receptor interaction pathway,systemic lupus erythematosus pathway,and renin-angiotensin system pathway;there were 7 co-expression genes:LOC317471,Olr462,Mcpt8I3, Olr718,Mcpt8I2,Cd80,and Ttn.②Comparing group Q with group M,group M with group K,there was one common expression pathway:the interaction pathway of neuroactive ligand-receptor;there were three co-expression genes:Cep295nl,Mcpt8I3, Fam64a.Conclusion:①The regulation of warming yang for resolving fluid retention therapy on renal medulla gene in rats with pleural effusion is the result of multiple pathways and gene interactions.②This therapy may affect the inflammatory response of Mcpt8I3 by up-regulating the interaction pathway of neuroactive ligand receptors.③There are differences in the pathway analysis of renal medulla gene between group W and group Q.Wenyang Xiaoyin Formula may play a diuretic and resolving fluid retention role by up-regulating Mcpt8I3 gene to regulate renin-angiotensin system pathway.④This method has no effect on the cAMP-PKA/PKC and VWP pathway in this study.
