摘 要:目的:探究补阳还五汤对脊髓损伤大鼠红核神经元L-型钙通道(ICa-L)电流及活动变化的影响。方法:将大鼠随机分为假手术组、模型组、补阳还五汤低、中、高剂量组,每组10只。采用红核脊髓束(rubrospinal tract,RST)横断方法,建立脊髓损伤动物模型,分离红核细胞。采用膜片钳技术记录各组ICa-L的变化及离子活动。结果:模型组红核细胞ICa-L电流密度在测试电压-20~+70 mV之间均明显低于假手术组(P<0.01),模型组曲线左移,ICa-L最大电流时的激活电压明显低于假手术组(P<0.01)。补阳还五汤低剂量组的ICa-L电流密度在测试电压20~+40 mV之间均高于模型组(P<0.05);补阳还五汤中、高剂量组的ICa-L电流密度在测试电压-10~+50mV之间均高于模型组(P<0.05或<0.01)。模型组ICa-L开放时间、开放概率均高于假手术组(P<0.05)。补阳还五汤中、高剂量组开放时间、开放概率均低于模型组(P<0.01)。结论:补阳还五汤治疗脊髓损伤可能与其抑制神经细胞ICa-L的开放防止钙超载有关。Objective:To study the effects of Buyang Huanwu Decoction (BYHWD)on L-type calcium channel (ICa-L)of Red Nucleus Neurons in rats with Spinal cord injury.Methods:The experiment rats were randomly divided into sham operation group,model group,BYHWD low groups,BYHWD mediate groups and BYHWD high groups,10rats in each group.The ruhrospinal tract was damaged in modal group and BYHWD group and given red nucleus neurons;the change of ICa-L currents were detected with Patch-clamp techniques. Results:Compared with the sham operation group,the model group showed lower ICa-L density of erythrocyte between test voltage -20mV and +70mV in Red Nucleus Neurons,and the curve of model group shifted to the left.The activation voltage at the maximum current of ICa-L was significantly lower than that of the sham operation group (P<0.01).The ICa-L current density in the BYHWD low dose group was higher than that of the model group between the test voltage 20mV and +40mV (P<0.05);The ICa-L current density of medium andhigh dose group in BYHWD was higher than that of the model group between the test voltage -10mV to + 50mV (P<0.05or P<0.01).Compared with the sham operation group,the ICa-L opening time of the model group was prolonged and the opening probability was increased (P<0.05).Compared with the model group,the opening time in the medium and high dose group of BYHWD was shortened and the opening probability decreased,which was statistically significant (P<0.01).Conclusion:BYHWD could inhibit L-type calcium channel in Red Nucleus Neurons of Spinal cord injury.It implied that BYHWD could inhibit L-type calcium channel in neuron.
