摘  要:目的 观察长链非编码RNA (lncRNA)牛磺酸上调基因1(TUG1)对人神经母细胞瘤侵袭和转移能力的影响.方法 神经母细胞瘤SK-N-SH细胞分为3组:TUG1小干扰RNA(si-TUG1)组、乱序对照小于扰RNA(si-NC)组和空白对照组.将TUGI-siRNA在阳离子脂质体Lipofectamine 2000的介导下,瞬时转染入神经母细胞瘤细胞株SK-N-SH,实时定量聚合酶链反应(Real-time PCR)检测3组细胞中TUG1表达水平;划痕实验和Transwell小室实验检测瘤细胞迁移和侵袭能力.结果 TUG1-siRNA转染神经母细胞瘤后,TUG1 mRNA表达下调.与对照组比较,si-TUG1组细胞迁移能力下降,细胞侵袭能力下降50.4.%(P=0.001).结论 TUG1在神经母细胞瘤细胞中高表达,且能促进瘤细胞的侵袭和转移能力.Objective To explore the effects of taurine upregulated gene 1 (TUG1) on migration and invasion of human neuroblastoma. Methods The cells were classified into threegroups: TUG1 small interfering RNA (si-TUG1) group, scramble control (si-NC) group, andblankcontrolgroup. Under the induction ofLipofectamine 2000, TUG1 small interfering RNA (TUG 1-siRNA) was transfected into neuroblastoma cells. The expression of TUG1 was detected by using real-time reversetranscriptionpolymerase chain reaction (Real-time PCR). The cell wound healing and transwellassays were applied for the detection of migration and invasion of cancer cells. Results Comparedtonegativecontrolgroup, the expressionof TUG1 was decreased. The cellular invasiveability was suppressed and metastatic activity was downregulated by 50.4% (P=0.001). Conclusion TUG1 is over expressed in neuroblastoma cells, andpromotes the migrationand invasionability of cancer cells.
