摘  要:目的探讨混合系列蛋白激酶样结构域(MLKL)在非小细胞肺癌(NSCLC)组织的表达水平及其与NSCLC患者临床病理学特征的关系,分析其与NSCLC患者生存的相关性。方法我院65例NSCLC患者手术新鲜标本,采用实时定量聚合酶联反应(Real-timePCR)检测NSCLC肿瘤组织和癌旁组织的MLKL的mRNA表达水平;分析MLKL的mRNA表达水平和NSCLC患者的基本临床病理学特征参数的关系;利用在线Kalpan-MeierPlotter网站分析癌组织中的MLKL表达水平和NSCLC患者生存的相关性。结果在NSCLC癌组织中MLKL的mRNA表达水平明显低于癌旁组织(1.00±0.13比5.69±0.49,P=0.000);NSCLC组织中MLKL的mRNA表达水平与T及N分期呈明显负相关(P=0.026、0.048),较低的MLKL的mRNA表达提示较差的组织分化(P=0.040);MLKL低表达的NSCLC总人群和腺癌人群的总生存期(OS)和进展后生存期(PPS)显著低于MLKL高表达组(P=0.000、0.006、0.014、0.014)。结论MLKL在NSCLC癌组织中显著低表达,与NSLCL的分化程度、T和N分期明显相关,其低表达提示预后较差。Objective To study the expression of mixed lineage kinase domain-like (MLKL) mRNA in non-small-cell lung carcinoma (NSCLC) tissue samples and analyze the correlation between MLKL and the clinicopathological features and survival in NSCLC. Methods Real-time quantitative polymerase chain reaction (Real-time PCR) was utilized to detect the expression levels of MLKL in cancer and adjacent tissues in NSCLC. The relationship between MLKL mRNA and the clinicopathological characteristics in NSCLC was analyzed. Online Kaplan-Meier database was used to explore the effects of MLKL on NSCLC overall survival (OS) and post-progression survival (PPS). Results The MLKL levels were significantly lower in NSCLC cancer tissues than those in adjacent tissues in NSCLC (1.00±0.13 vs. 5.69±0.49, P=0.000). MLKL levels were correlated to T stage and N stage negatively (P=0.026, 0.048). OS and PPS of NSCLC patients with lower levels of MLKL were significantly shorter than those with higher levels of MLKL expression (P=0.000, 0.006, 0.014, 0.014). Conclusion MLKL was decreased in NSCLC cancer tissues, and related to differentiation positively, and to tumor size and lymph node metastasis negatively. Decreased expression of MLKL is associated with shorter survival in NSCLC.
