摘  要:目的:探讨趋化因子CCL2(Chemokine C-C motif Ligand 2)对小鼠黑质多巴胺能神经元的损伤。方法:将CCL2单次注射入C57BL/6J小鼠黑质部位,在注射前和注射后的各个时间点分别记录小鼠的行为轨迹,并将注射后不同时间点的小鼠灌注固定取脑,通过免疫组织化学方法检测黑质多巴胺能神经元的数量变化。结果:免疫组化检测显示在CCL2注射后21d开始黑质多巴胺能神经元的数量明显减少,并显著低于对照组,一直持续到注射后28d;并且在注射后28d时小鼠的运动速度显著低于对照组。结论:脑内注射CCL2可以损伤小鼠黑质多巴胺能神经元。Objective: To study the effects of chemokine CCL2 on dopaminergic neurons in substantia nigra ( SN) of mice.Methods: A single intronigral injection of Chemokine C-C motif Ligand 2 ( CCL2) was made.At certain time points,the behavioral trajectories of mice were recorded,and the brain was perfused and fixed at different time points after injection.The number of dopaminergic neurons in substantia nigra was detected by immunohistochemistry.Results: Immunohistochemistry showed that the number of dopaminergic neurons in substantia nigra decreased significantly from 21 days after CCL2 injection compared with the control group,decreasing continuously till 28 days.Moreover,the movement speed of mice was significantly lower than that of the control group on the 28 days after CCL2 injection.Conclusion: Intranigral injection of CCL2 can damage dopaminergic neurons in mouse substantia nigra.
