野黄芩苷对骨肉瘤细胞系MG-63的增殖侵袭的抑制作用和Bax/Bcl-2表达的影响

摘　　要：目的研究野黄芩苷(scutellarin)对骨肉瘤细胞系MG-63的增殖侵袭和Bax/Bcl-2表达的影响。方法将骨肉瘤细胞系MG一63做4组处理:MG-63对照组(MG-63),野黄芩苷低、中、高剂量组(10、20、50μmoL/L);细胞计数试剂盒(cell counting kit-8,CCK8)检测细胞增殖;流式细胞术检测细胞凋亡情况;Transwell检测细胞侵袭能力;蛋白质印迹检测Ki67、半胱氨酸天冬氨酸特异性蛋白酶3(cvsteine aspartate specific protease-3,Caspase-3)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达;水溶性四氮唑法检测总超氧化物歧化酶(superoxide dismutase,SOD)的活力和硫代巴比妥酸法检测丙二醛(malondialdehyde,MDA)的含量;蛋白质印迹检测Bax和B细胞淋巴瘤2(B-cell lymphoIna-2,Bcl-2)的表达。结果MG-63细胞经野黄芩苷加药处理后,细胞增殖速率显著降低;凋亡细胞的比率显著上升;细胞侵袭能力显著降低;Ki67的表达显著下调,Caspase-3的表达显著上调,VEGF的表达显著下调;总SOD的活力显著降低,MDA的含量显著上升;Bax的表达显著上调,Bcl-2的表达显著下调。结论野黄芩苷抑制骨肉瘤细胞系MG-63的增殖侵袭,提高Bax/Bcl-2的比例。Objective To investigate the effect of scutellarin on proliferation,invasion and Bax/Bel-2expression of osteosarcoma cell line MG-63.Methods Osteosarcoma cell line MG-63 was divided into 4groups:MG-63control group,low-,medium-and high-dose scutellarin groups (10,20,50μmoL/L Scutellarin,respectively).CCK8was used to detect cell proliferation;flow cytometry was used to detect cell apoptosis;Transwell was used to detect the cell invasion ability; Western blotting was used to detect expression of Ki67,cysteine aspartic proteinase 3(Caspase-3),vascular endothelial growth factor (VEGF),Bax and Bel-2;related kits were used to detect the activity of total SOD and the content of MDA Results After treatment with baicalin,the per-centage of apoptotic cells,the expression of Caspase-3,and the content of MDA was increased significantly ;proliferation rate of the MG-63cells,the cell invasiveness,the expression of Ki67,the expression of VEGF,and the activity of the total SOD decreased significantly.The expression of Bax was significantly up-regulated and the expression of Bcl-2significantly down-regulated.Conclusion Scutellarin inhibits proliferation and invasion of osteosarcoma cell line MG-63and increases the proportion of Bax/Bcl-2.
