摘 要:目的利用高脂喂养结合大脑中动脉栓塞法建立痰瘀互结证局部脑缺血再灌注动物模型,以验证该造模方法的可行性。方法将35只雄性SD大鼠按随机数字表法分为普通喂养组15只和高脂喂养组20只,分别予以普通饲料和高脂饲料喂养4周。再将大鼠重新编号,普通喂养组随机分为正常组10只和普通模型组5只,高脂喂养组随机分为假手术组10只和高脂模型组10只,其中两个模型组采用大脑中动脉栓塞法复制脑缺血再灌注模型。造模完成24 h后进行Zea Longa评分以评价其神经功能缺损程度;继续喂养1周后,采血检测血脂四项(TC、TG、LDL-C、HDL-C)及凝血功能(APTT、PT、FIB)水平,取脑计算脑系数及制作脑组织病理切片,以评价模型的建立和病理生理特点。结果经过4周的喂养,与普通喂养组比较,高脂饲料喂养组大鼠日平均饮食量及饮水量均明显减少,差异有统计学意义(P<0.05)。喂养10天、20天后,高脂喂养组大鼠体重低于普通喂养组(P<0.05),喂养4周后,两组体重比较差异无统计学意义(P>0.05)。完成造模后,模型组左侧肢体偏瘫,右侧面瘫症状明显,行走呈逆时针追尾状,甚或往健侧倾倒,精神萎靡,活动减少,身体蜷缩,舌质紫暗。与正常组、普通模型组比较,高脂模型组TC、TG、LDL-C、HDL-C水平均显著升高(P<0.05);与正常组比较,两模型组APTT、PT值显著降低(P<0.05),高脂模型组脑系数值升高(P<0.05);与假手术组比较,高脂模型组PT值降低(P<0.05)、脑系数升高(P<0.05)。两模型组间APTT、PT、FIB及Zea Longa评分比较,差异均无统计学意义(P>0.05)。结论高脂饲料喂养结合大脑中动脉栓塞法在SD大鼠体内可以成功建立病证结合的痰瘀互结证局部脑缺血再灌注模型。Objective To establish the animal model of focal cerebral ischemia/reperfusion with intermingled phlegm and blood stasis syndrome(IPBSS) by high-fat feeding combined with middle cerebral artery occlusion(MCAO), thus verifying the feasibility of this modeling method. Methods Thirty-five male SD rats were randomly divided into the normal feeding group(n=15) and the high-fat feeding group(n=20) using random number table. Those in the normal feeding group were fed with ordinary food, while those in the high-fat feeding group were fed with high-fat food. All feed lasted for 4 successive weeks. And then rats in the normal feeding group were re-randomized into a normal group(n=10) and an ordinary model group(n=5), and rats in the high-fat feeding group were re-randomized into a sham-operated group and a high-fat model group, 10 in each group. The focal cerebral ischemia/reperfusion model was duplicated in those in the two model group by MCAO. Zea Longa score was implemented 24 h after modeling to evaluate the degree of nervous functional defects. The feeding was continued for 1 week. Blood lipids(TC, TG, LDL-C, HDL-C) and coagulation functions(APTT, PT, FIB) were measured. Brain coefficients were calculated and histopathological sections of brain tissues were prepared to evaluate the establishment of the model and the pathophysiological characteristics. Results After 4 weeks of feeding, the average daily food intake and drinking water amount of rats were significantly reduced in the high-fat feed group, with statistical difference as compared with the normal feeding group(P<0.05). Afte 10 days and 20 days of feeding, the weights of rats in the high-fat feeding group were lower than those in the normal feeding group(P<0.05), and after 4 weeks of feeding, there was no difference in weight between the two groups(P>0.05). After modeling, left limbs were paralyzed and right facial paralysis occurred obviously in the model group. They walked counter-clockwisely tail-tracking, and they were listless.
