摘  要:7,8-二羟基黄酮(7,8-DHF)是天然存在的黄酮类化合物家族成员之一。研究表明,7,8-DHF能够穿过血脑屏障,有效模拟脑源性神经营养因子(BDNF),专一性地结合TrkB受体,从而诱导TrkB发生二聚化及自磷酸化,并进一步激活受体下游的MAPK/ERK、PI3K/Akt以及PCγ1三条信号通路。目前,7,8-DHF被应用于各类BDNF/TrkB信号相关疾病的防治研究,并取得一系列令人振奋的成果。本文在简要介绍BDNF及其受体TrkB的基础上,综述了7,8-DHF激活TrkB受体的分子机制及其在各类相关疾病模型(如阿尔兹海默症、帕金森症、亨廷顿舞蹈症、Rett综合症、抑郁症及肥胖等)中的防治效果研究情况,以期为黄酮类化合物作为神经性疾病防治及其体重控制的功能性食品的开发提供参考。7,8-dihydroxyflavone (7,8-DHF)is one of the members of flavonoids family which exist in nature as a sort of important bioactive compounds.Recently,7,8-DHF was found to potently mimic brain-derived neurotrophic factor (BDNF),one of the most important neurotrophins inside of our body,to selectively bind tropomyosin-related kinase receptor B (TrkB),and then stimulate TrkB dimerization and autophosphorylation,thus resulting in the activation of three major downstream signaling pathways involving MAPK/ERK,PI3K/Akt and phospholipase Cγ1.Therefore this significant physiological function of 7,8-DHF received extensive attention.Nowadays 7,8-DHF has been applied to the research on various diseases related to BDNF/TrkB signaling pathways and many encouraging results have been reported.In order to provide evaluable information on the study and development of flavonoids to be as a kind of functional food in prevention of some neurological diseases and weight management,this paper here overviews the recent work on 7,8-DHF as a selective TrkB agonist,including the mechanism of 7,8-DHF binding to TrkB receptor and its effects on BDNF/TrkB signaling-related diseasses,like Alzheimer's Disease,Parkinson's Disease,Huntington Disease,Rett Sydrome,depression and obesity,based on the brief introduction of BDNF and its receptor TrkB.
