PAK6在膀胱癌中表达及其临床意义

摘　　要：目的探讨p21活化激酶6(p21一activated kinase 6,PAK6)在膀胱癌中的表达情况及其临床意义。方法膀胱癌患者89例(膀胱癌组),取其膀胱癌组织;行膀胱部分切除或全部切除术患者30例为对照组,取其正常膀胱上皮组织。采用免疫组织化学法检测2组组织PAK6蛋白阳性表达率。Biu-87、T24膀胱癌细胞和SV-HUC-1正常膀胱细胞分为Biub一87组、T24组和SV-HUC-1组,采用实时荧光定量PCR法检测3组细胞PAK6mRNA表达水平,采用Westernblot法检测3组细胞PAK6蛋白表达水平。结果膀胱癌组PAK6蛋白阳性表达率(46.07%)明显低于对照组(96.67%)(P<O.05),膀胱癌组复发者PAK6蛋白阳性表达率(29.41%)低于原发者(56.36%)(P<O.05),膀胱癌组PAK6蛋白阳性表达率在肿瘤数量、病理分级、是否转移和临床分期上差异无统计学意义(P>O.05);Biu一87组和T24组细胞PAK6mRNA(0.0189±0.0037、0.053O±0.0129)和PAK6蛋白表达水平(O.0993±0.0121、0.1431±0.0081)明显低于SV-HUC-1组(1.0403±0.3755、0.3381±0.0511),Biu-87组低于T24组(P<O.05)。结论PAK6在膀胱癌组织及细胞中呈低表达,且与膀胱癌的复发密切相关,可能在膀胱癌的发生、发展中起抑制作用,可作为新的抑癌指标指导临床。Objective To investigate the expression and significance of p21-activated kinase 6(PAK6)protein in bladder cancer.Methods Immunohistochemistry was used to analyze the expression of PAK6protein in the bladder cancer tissue from 89patients with bladder cancer (cancer group)and the normal epithelial tissue from 30 patients undergoing total or partial cystectomy (control group).The expressions of PAK6mRNA and protein were detected by real-time fluorescence quantitative PCR and Western blot method respectively in Biu-87ceils,T24cells and SV-HUC-1cells.Results The positive expression rates of PAK6protein were significantly lower in cancer group (46.07%)than that in control group (96.67%)(P<0.05),and in patients with recurrent cancer (29.41%)than that in patients with primary cancer (56.36%)(P<0.05),while no significant differences were found in the number,pathological grade,metastasis and the clinical stage of bladder cancer (P>0.05).The positive expression rates of PAK6mRNA and protein increased gradually inBiu-87cells (0.0189±0.0037,0.0993±0.0121),T24cells (0.0530±0.0129,0.1431±0.0081)and SV-HUC-1cells (1.0403±0.3755,0.3381±0.0511)(P<0.05).Conclusion PAK6protein is lowly expressed in bladder cancer,and is closely correlated with the recurrence of bladder cancer,playing an inhibitory role in the development and progress of bladder cancer,therefore it might be used as a new antitumor marker.
