摘  要:本研究采用吹扫捕集,结合气相色谱-质谱联用技术,建立了一种测定人体尿液中丙酮含量的新方法。该方法对水体中丙酮的检出限为13.18μg/L,在20~1 000μg/L浓度范围内线性关系良好,相关系数R^2>0.99,相对标准偏差为6.73%(n=5)。将该方法应用于实际人体尿液中丙酮含量的测定时,选择201-二甲基硅油作为尿液消泡剂,加标回收率在87.0%~98.0%之间。通过比较糖尿病病人和正常人尿液中的丙酮含量,发现尿液中丙酮浓度和血糖值之间存在明显的正相关性,相关系数达到0.998,说明该方法在糖尿病病情监测及诊断方面有较好的应用前景。The acetone content in human urine might provide a reliable platform for the diagnosis and classification of diabetes mellitus because the level of acetone in diabetics is higher than that in normal people,Herein,an automated method was established for the detection of acetone content in human urine using purge-and-trap coupled to gas chromatography with mass spectrometry.Under the optimized conditions,the analytical method provided a detection limit of 13.18μg/L with a good linear correlation(R^2>0.99)and precision(relative standard deviation less than 6.73%(n=5)).Satisfactory recoveries of87%-98% were obtained for the human urine samples when 201-simethicone was used as defoaming agent.By comparing the content of acetone in the urine of diabetics with that of normal people,it was found that the concentration of acetone in urine showed a significant positive correlation(correlation coefficient>0.998)with blood glucose concentration.These results indicate the proposed method has great practical value for the diagnosis and monitoring of diabetes mellitus.
