摘 要:目的观察姜黄素对人恶性胶质瘤细胞体外生长及血管生成拟态的影响及姜黄素的干预作用。方法培养细胞,普通显微镜下观察姜黄素对U87MG细胞血管生成拟态的影响,MTT法筛选姜黄素最佳浓度后将其分为4组,采用免疫细胞化学技术、WestenBlot技术检测相关蛋白的表达情况,PCR技术检测基因表达变化。结果人恶性胶质母细胞瘤细胞体外培养时可发生血管生成拟态现象,姜黄素干预后血管生成拟态现象不明显,VE-cadherin/EphA2/PI3K/MMP-2细胞信号转导通路中各因子蛋白和mRNA的表达下降。结论姜黄素可能通过影响VE-cadherin/EphA2/PI3K/MMP-2细胞信号转导通路中各因子干预血管生成拟态现象,从而达到抑制胶质母细胞瘤生长的作用。Objective To observe the effect of curcumin on growth and angiogenesis mimicry of human malignant glioblastoma cells in vitro and the intervention effect of curcumin. Methods The cells were cultured and the effect of curcumin on the angiogenesis mimicry of U87 MG cells was observed under normal microscope. The optimal concentration of curcumin was screened by MTT method and divided into 4 groups. The expression of related proteins was detected by immunocytochemistry and WestenBlot, PCR technique to detect changes in gene expression. Results The angiogenesis mimicry could occur in human malignant glioblastoma cells in vitro. The angiogenic mimicry was not obvious after curcumin intervention. Each factor proteins in cell signal transduction pathways of VE-cadherin/EphA2/PI3 K/MMP-2 and the expression of m RNA decreased. Conclusion Curcumin may inhibit the growth of glioblastoma by affecting the angiogenesis mimicry in various factors in the signal transduction pathway of VE-cadherin/EphA2/PI3 K/MMP-2.
