摘 要:目的将Fluoro-Ruby和Fluoro-Gold运用于大鼠坐骨神经慢性卡压模型,并探讨大鼠坐骨神经慢性卡压后Fluoro-Ruby染色的背根神经元是否能将轴突受损的背根神经元从众多背根神经元中分辨出来。方法将16只普通级SD雄性大鼠随机分成两组,每组8只,A组按照Mackinnon设计的方法建立坐骨神经慢性卡压模型,B组则仅暴露坐骨神经;术后2周,将5%Fluoro-Ruby和5%Fluoro-Gold各5μL分别注射卡压段神经的近端和远端。1周后,分别取A、B两组大鼠右侧L4、L5、L6背根神经节,行冰冻切片后立即在荧光显微镜下观察Fluoro-Ruby和Fluoro-Gold染色的背根神经元并照相保存。结果 Fluoro-Ruby和Fluoro-Gold可以分别对不同的背根神经元进行染色,B组大鼠背根神经节内可见(18.2±1.5)个Fluoro-Ruby示踪染色的背根神经元,而A组大鼠背根神经节内可见(38.1±4.5)个Fluoro-Ruby示踪染色的背根神经元细胞,数目较B组多,差异有统计学意义(P<0.05);A组背根神经节内可见(124.3±8.4)个Fluoro-Ruby和Fluoro-Gold示踪染色的背根神经元细胞,与B组的(122.6±4.7)个染色神经元细胞数目大致相同,差异无统计学意义(P>0.05)。结论 Fluoro-Ruby和Fluoro-Gold示踪染色背根神经节运用于大鼠坐骨神经慢性卡压后,Fluoro-Ruby染色的背根神经元可以将轴突受损的背根神经元从众多背根神经元中辨别出来。Objective To investigate the application of Fluoro-Ruby and Fluoro-Gold retrogradely labeled neu-rons of dorsal root ganglion in chronic sciatic nerve compression in rats, and to explore whether the neurons labeled byFluoro-Ruby can identify the injured neurons from numerous neurons. Methods A total of 16 male Sprague-Dawleyrats were randomly divided into two groups: rats in group A (n=8) subjected to sciatic nerve compression operation ac-cording to the method described by Mackinnon, and rats in group B (n=8) had sciatic nerve exposed only. Ipsilateralnerves of rats from groups A and B were again exposed for injection with 5μL of 5% Fluoro-Gold and 5% Fluoro-Rubysolution 2 weeks after surgery. Fluoro-Gold was injected into the nerve distal to the region occupied by Silastic tubing,while Fluoro-Ruby was infused proximally. L4/L5/L6 ipsilateral dorsal root ganglions were harvested 3 weeks post injury.dorsal root ganglions-containing axonal tracers were sectioned on a cryostat at a thickness of 10 μm, and then sectionswas observed with a fluorescence microscope. Results The number of neurons with Fluoro-Ruby was (18.2±1.5) ingroup B, significantly less than (38.1±4.5) in group A (P<0.05). The total number of neurons with Fluoro-Ruby and Fluo-ro-Gold was (124.3±8.4) in group A, versus (122.6±4.7) in group B (P>0.05). Conclusion After the applying of Fluo-ro-Ruby and Fluoro-Gold retrogradely labeled neurons of dorsal root ganglion to chronic sciatic nerve compression, Flu-oro-Ruby labeled neurons can help distinguish injured neurons from numerous neurons in dorsal root ganglion.
