摘 要:目的优化太白米总黄酮超声提取工艺,并测定其体外抗氧化能力。方法以乙醇体积分数、料液比、提取时间、提取温度、提取功率和提取次数为影响因素,总黄酮得率为评价指标,在单因素试验基础上,运用响应面法探讨乙醇体积分数、料液比、提取时间和提取温度对太白米总黄酮得率的影响并进行优化;评价其清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2-联氮基双-3-乙基苯并噻唑啉-6-磺酸(ABTS+)的能力以及对铁离子(Fe3+)的还原能力,并以抗坏血酸为阳性对照。结果最佳工艺为乙醇体积分数62%,料液比1∶40(m L/g),提取时间32 min,提取温度70℃,总黄酮得率为15.16 mg·g-1;当醇提液总黄酮含量为50μg·mL-1时,其DPPH自由基清除率、ABTS+自由基清除率和FRAP的OD值分别为86.68%、86.43%和0.73,抗氧化能力较强。结论该方法可为太白米总黄酮的提取和应用提供一定科学依据。Objective To optimize the ultrasound-assisted extraction process of total flavonoids from Notholirion bulbuliferum,and to measure the in vitro antioxidant activities of the extracted total fl/tvonoids. Methods The effect of ethanol concentration,solid-liquid ratio,extraction time,extraction temperature, extraction power and extraction times on extraction efficiency of total flavonoids was analyzed by a single factor test,The optimum extraction process of total flavonoids from Notholirion bulbuliferum was analyzed by Box-Behnken response serface.The antioxidant activities of the total flavonoids were detected by clear 1, 1-diphenyl-2-trinitrophenylhydrazine (DPPH)radicals,2,2'-nitrilobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS +)and the reducing ability of Fe^3+and ascorbic acid was regarded as the positive control.Results The optimal ultrasound-assisted extraction conditions for total flavonoids from Notholirion bulbuliferum were as follows:ethanol concentration 62%,liquid-solid ratio at I :40(mL/g),extraction for 32min,and extraction temperature at 70℃.Under these conditions,the yield of total flavonoids was 15.16mg·g^-1.The extracted flavonoids exhibited powerful in vitro antioxidant activities and its scavenging rate of DPPH,ABTS +and OD value of FRAP were 86.68%,86.43%and 0.73,respectively.Conelusion This study provides a theoretical basis for further development and utilization of total flavonoids from Notholirion bulbuliferum.
