摘  要:目的研究注射用鼠神经生长因子(mNGF)对六溴环十二烷(HBCD)暴露下小鼠脑损伤的保护作用。方法 2月龄昆明小鼠分为正常对照组、HBCD组和mNGF组。mNGF组小鼠腹腔注射mNGF 5ng/g·d,连续5d。然后,对HBCD组和mNGF组小鼠每天灌入溶有HBCD的花生油100μl(浓度为50μM/ml),正常对照组每天灌入等量不含HBCD的花生油,连续10d。采用试剂盒及ELISA方法检测小鼠脑组织中相关酶活性如乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD),谷胱甘肽(GSH),丙二醛(MDA)及凋亡相关蛋白Bcl-2。结果小鼠暴露于50μM/ml HBCD 10d后,LDH释放增加,SOD和GSH酶活性降低,MDA含量增加,Bcl-2含量下降。mNGF能够显著提高抗氧化酶的活性及凋亡相关蛋白Bcl-2的表达,降低MDA的活力。结论 m NGF对小鼠暴露HBCD造成脑损伤的保护作用可能与抗凋亡相关。Objective To investigate the protective effect of mouse nerve growth factor(mNGF) on mice brain damage caused by hexabromocyclododecane(HBCD). Methods Two months old Kunming mice were divided into control group, HBCD group and mNGF(intraperitoneal injected mNGF 5 ng/g daily for 5 days) group. Then, HBCD group and mNGF group mice were fed with peanut oil 100μl containing HBCD(50μM/ml) every day by gavage for 10 days, the control group was fed with peanut oil without HBCD. The related enzyme activities were detected in brain tissue, such as lactate dehydrogenase(LDH), superoxide dismutase(SOD), glutathione(GSH) and malondialdehyde(MDA);the apoptosis related protein Bcl-2 was detected by ELISA. Results The content of LDH was increased, enzyme activities of SOD and GSH decreased, content of MDA increased, but content of Bcl-2 decreased after mice were exposured to 50μM/ml HBCD for 10 d. However, mNGF significantly upregulated the activites of enzymes and the level of apoptosis related protein Bcl-2, and downregulated the activity of MDA. Conclusion The protective effect of m NGF on mice brain damage caused by HBCDmight be related to the anti-apoptosis effect.
