摘 要:目的本文主要讨论MMP-9抑制剂对大鼠脑出血模型脑水肿的影响及其对血脑屏障的影响情况。方法将成年雄性Wistar大鼠分为对照组、实验组、用药组,对照组共用7只大鼠,实验组及用药组各28只大鼠,实验组及用药组分别于12h、24h、3d、5d等时间点分成4个亚组,每个亚组7只大鼠。其中实验组做脑出血模型,用药组做脑出血模型后注射MMP-9拮抗剂,其他组注射等量的生理盐水。统计分析对照组、实验组、用药组及其亚组之间脑水肿差别、血脑屏障破坏的差别、血肿周围水肿凋亡细胞的差别、血肿周围水肿带β-DG表达的量及形式的差别。结果用药组的脑神经细胞排列稍显紊乱,存在一定程度的结构异常及核固缩和胞质凝集现象,但程度均明显低于实验组,用药组脑组织含水量明显低于实验组,用药组脑组织细胞凋亡数明显低于实验组,差异具有统计学意义(P<0.05);用药组血脑屏障通透性明显低于实验组,以上差异均具有统计学意义(P<0.05)。结论 MMP-9抑制剂对大鼠脑出血后水肿、神经组织损伤有抑制作用,对血脑屏障有一定的保护作用。Objective To discuss the effect of Cerebral Edema on Cerebral Hemorrhage and the role of the blood- brain barrier in Rats by MMP-9inhibitor.Methods The adult male Wistar rats were divided into control group,experi- mental group and drug group.The control group shared 7rats.The experimental group and the drug groul5each had 28 rats.The experimental group and drug group were at 12h,24h,3d,5d respectively At equal time points were divided into four subgroups,7 rats in each.subgroup.In the experimental group,intracerebral hemorrhage models were per- formed.In the medication.group, intracerebral hemorrhage models were used to inject MMP-9antagonists,and other' groups were injeeted with equal volume of physiological.saline.Statistical analysis of differences in brain edema,differ- ence in blood-brain barrier disruption between control group,experimental group,medication group,and subgroups, difference in apoptotic cells around hematoma edema,and amount and form of β-DG expression around edema around hematoma.Results The arrangement of cerebral nerve.cells in the medication group was slightly disordered,with a cer- tain degree of structural abnormalities and nuclear condensation and cytoplasm agglutination,butt.he degree was signifi- cantly lower than that of the,experimental group.The difference was statistically significant (P<0.05);The tissue wa- ter content was significantly lower than that of the experimental group,and the difference ,was statistically significant (P<d.05).The number of apoptotic'cells in the brain of the drug.group,was significantly lower than that of the experi- mental group (P<0.05);the blood brain'barrier was dsed in the drug group.Permeability was significantly lower than that of the experimental group,and the difference was statistically significant (P<0.05).Conclusion MMP-9inhibi- tors have inhibitory effects on edema and nerve tissue damage after intracerebral hemorrhage in rats,and have a protec- tive effect on the blood-brain barrier.
