摘　　要：依米珠单抗是人源化的单克隆抗体,可替代失活的因子Ⅷ(FⅧ)在血浆中与活性因子Ⅺ(FⅪ)和因子Ⅹ(FⅩ)结合发挥止血作用。依米珠单抗半衰期长,体内止血作用不受FⅧ抑制物的影响。2017年11月16日美国食品和药物管理局批准依米珠单抗注射液用于存在FⅧ抑制物的成人和儿童血友病A的常规预防治疗及减少出血发作的频次,推荐用法为前4周依米珠单抗3 mg·kg^(-1)每周皮下注射1次, 4周后1.5 mg·kg^(-1)每周1次。依米珠单抗引起的主要不良反应为注射部位反应、头痛、疲劳、上呼吸道感染及关节痛等。Emicizumab is a bispecific humanized monoclonal antibody that restores the function of missing activated factor Ⅷ (F Ⅷ )by bridging activated factor Ⅸ and factor Ⅹ for effective hemostasis. Emicizumab has long half-life and is effective regardless of FⅧ inhibitors.On November 16,2017,the U.S. Food and Drug Administration approved emicizumah injection for routine prophylaxis to prevent or reduce the frequency of bleeding episodes in adults and pediatric patients with hemophilia A with F Ⅷ inhibitors.The recommended dose is 3 mg·kg^-1 by subcutaneous injection once weekly for the first 4 weeks,followed by 1.5 mg·kg^-1 once weekly.The most common adverse reactions are injection site reaction,headache,fatigue,upper respiratory tract infection,joint pain,and so on.
