摘 要:目的明确硒代蛋氨酸(Se-Met)对慢性低氧大鼠缺氧诱导因子1α(HIF-1α)、血管内皮生长因子(VEGF)的影响,及对心肌细胞线粒体结构的影响。方法将SD大鼠共分为9组,设常氧组1组,海拔3 000 m和5 000 m各4组:低氧对照组,低氧1、2、3补硒组,补硒组饲料中加入Se-Met,分别为参考摄入量的5、10、20倍。低氧模拟实验在低压氧舱进行(30d)。用ELISA法测定各组大鼠血浆HIF-1α、VEGF水平;用电镜观察心肌细胞线粒体改变。结果在海拔3 000 m和5 000 m的低氧对照组的HIF-1α水平高于补硒各组,HIF-1α水平随补硒量的增加而降低,5 000 m补硒各组下降明显。VEGF水平在海拔3 000 m组均低于常氧组,VEGF水平随补硒量增加而降低。补硒组大鼠心肌细胞线粒体的形态明显接近于常氧对照组。结论 Se-Met对慢性低氧大鼠HIF-1α、VEGF水平存在影响,能够减轻低氧对心肌细胞线粒体结构的影响。Objective To investigate the effect of Se-Met on hypoxia induce factor-1 alpha (HIF-1 α)and vascular endothelial growth factor(VEGF)and mitochondrial structure of myocardial cells in chronic hypoxic rats at different altitude.Methods SD rats were divided into 9 groups,including 1 normoxic group,4 groups with altitudes of 3 000 m and 5 000 m(hypoxic control group,hypoxic 1,2 and 3 selenium supplementation groups),and Se-Met were added into the feed of selenium supplementation groups,which were 5,10 and 20 times the reference intake, respectively.And the hypoxic simulation experiment were carried out in the low-pressure oxygen chamber 30 days. HIF-1α and VEGF levels were determined by ELISA.Mitochondrial changes of myocardial cells were observed by electron microscopy.Results The HIF-1α level in the hypoxic control groups at an altitude of 3 000 m and 5 000 m were higher than that in the selenium supplementation groups.The HIF-1α level decreased with the increase of selenium supplementation,and the decrease were significant in the selenium supplementation group at 5 000 m.VEGF level in the 3 000 m groups were lower than that in the normoxic groups.VEGF level decreased with the increase of selenium supplementation.And the myocardial cells mitochondria morphology of selenium supplementation group was remarkably similar with the normoxic control groups.Conclusions Se-Met may have effects on HIF-1α and VEGF levels in chronic hypoxic rats,and it could reduce mitochondrial structure of the myocardial cells under the condition of hypoxia.
