摘  要:目的探讨异基因骨髓移植联合胸腺上皮细胞移植动物模型的T细胞重建情况、抗肿瘤效应和移植物抗宿主病(GVHD)程度。方法采用异基因骨髓内骨髓移植及胸腺内胸腺上皮细胞移植方法,移植后观察受鼠GVHD临床表现并评分,移植后2周于受鼠右下背部种植肿瘤细胞并观测肿瘤大小至荷瘤后第4周,移植后4周流式细胞仪检测受鼠脾脏中T细胞亚群比例和细胞因子分泌情况,并对受鼠肝、小肠、皮肤组织行GVHD病理评分。结果异基因骨髓移植联合胸腺上皮细胞移植组(实验组)受鼠脾脏中CD4^+和CD8^+T细胞比例均高于异基因骨髓移植组(对照组),实验组受鼠脾淋巴细胞中可见胞内因子γ干扰素(INF-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)阳性细胞分群;荷瘤后实验组的肿瘤增长最慢,肿瘤体积最小;各组受鼠均无明显的GVHD表现。结论异基因骨髓移植联合胸腺上皮细胞移植可促进移植后的T细胞重建、增强抗肿瘤效应,同时并不引起明显的移植物抗宿主病。Objective To investigate the T -cell reconstitution,anti-tumor effect and the degree of graft -versus-host disease (GVHD)in animal model of allogeneic bone marrow transplantation plus thymic epithelial cell transplantation.Methods Allogeneic bone marrow transplantation plus intrathymic thymic epithelial cell transplantation were carried out.GVHD clinical manifestations of recipient mice were observed after transplantation.Two weeks after transplantation,tumor cells were implanted on the lower right back of recipient mice.Tumor size was observed until the fourth week of tumor -bearing.Four weeks after transplantation,the proportions of CD4^+ and CD8^+T cell,and cytokine secretion in the spleen cells of recipient mice were detected by flow cytometry;and histopathology of liver,intestine,skin were examined to assess pathological degrees of graft -versus -host disease.Results CD4^+and CD8^+T cell proportions of the allogeneic bone marrow transplantation combined with thymic epithelial cell transplantation group (Experimental group) were significantly higher than those of allogeneic bone marrow transplantation group (Control group).Cytokine INF -γ, TNF-α,IL-2 positive cells could be detected in the spleen lymphocytes of experimental group recipient mice.The tumor growth of experimental group was slower with smaller size.And there was no evidence of graft -versus-host disease in all recipient mice.Conclusion Allogeneic bone marrow transplantation combined with thymic epithelial cell transplantation can promote T-cell reconstitution,and enhance the anti-tumor effect without obvious GVHD after transplantation.
