摘 要:[目的]利用S-180荷瘤小鼠及细胞研究黄连素抑制肿瘤的作用及分子机制。[方法]利用免疫组织化学染色、WST1实验、流式细胞术及Real-timePCR等技术,检测黄连素抑制S-180肉瘤细胞的作用及机制。[结果]体内实验结果表明黄连素组生存时间明显延长(P<0.05),抑瘤率为25.19%,肿瘤直径(2.11±0.39)显著小于对照组(2.48±0.20),磷酸化组蛋白H3及CyclinD1阳性细胞数目明显降低(P<0.05)。体外实验结果表明黄连素可以抑制S-180细胞的生长,并诱导凋亡(P<0.05),Real-timePCR结果表明,黄连素组C-myc、CyclinD1、Bcl-2及FAS表达水平降低,Caspase-1及PGC-1α表达水平升高(P<0.05)。[结论]黄连素在体内水平抑制S-180荷瘤小鼠肿瘤的增殖,延长生存时间,在体外水平抑制S-180细胞的生长并诱导凋亡,黄连素通过调控增殖、凋亡及代谢等相关基因发挥抗肿瘤作用。[Objective]To study the inhibitory effect and molecular mechanisms of berberine on S-180 tumor-bearing mice and cells.[Methods]Immunohistochemical staining,WST1 cytotoxicity test,flow cytometry and Real-time PCR were used.[Results]The results of in vivo experiment showed that the survival time of the berberine group was significantly prolonged(P<0.05),the tumor inhibition rate was 25.19%,the average diameter of tumor(2.11 ± 0.39)was significantly lower than that of the control group(2.48 ± 0.20),and the number of phosphorylated histone H3 and Cyclin D1 positive cells was significantly decreased(P<0.05).The results of in vitro experiments showed that berberine could inhibit the growth of S-180 cells and induce apoptosis(P<0.05).Real-time PCR showed that the expressions of C-myc,Cyclin D1,Bcl-2 and FAS were decreased,while the expressions of Caspase-1 and PGC-1a were increased in berberine-treated group(P<0.05).[Conclusion ]Berberine could inhibit the proliferation of S-180 tumors in vivo,and prolong the survival time of S-180 tumor-bearing mice.Berberine could inhibit the growth of S-180 cells and induce apoptosis in vitro.Berberine plays an anti-tumor role by regulating proliferation,apoptosis and metabolism related genes.
