摘 要:目的:分析探讨H19基因rs2839698 C> T位点单核苷酸多态性与中国汉族人群口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)易感性的关系,并分析吸烟和饮酒与OSCC发病风险的关系。方法:采用PCR-RFLP基因分型方法对124例经病理诊断为新发OSCC患者和121例性别年龄均匹配的对照的rs2839698位点进行基因型检测,运用χ2检验和单因素、多因素Logistic回归等分析其与OSCC发生的相关性。结果:OSCC患者较对照人群的吸烟与饮酒比例显著增高;关联研究结果显示rs2839698位点CT基因型携带者较CC基因型会显著增加OSCC发病风险,且显性模型、隐性模型和超显性模型结果均显示存在边缘显著关联;分层分析结果显示,在女性、年龄≥60岁、吸烟人群和不饮酒人群具有更为显著的统计学相关性。结论:rs2839698 C> T位点多态性改变可能与中国汉族人群OSCC的发生相关;长期吸烟和饮酒可能会增加OSCC的发病风险。Objective: To investigate the relationship between the rs2839698 polymorphism in H19 gene and the susceptibility of oral squamous cell carcinoma,and to analyze the association between tobacco and alcohol intake with the risk of OSCC. Methods: PCR-RFLP method was used to genotype the rs2839698 polymorphism in one age-and sex-matched casecontrol study including 124 oral squamous cell carcinoma( OSCC) cases and 121 cancer-free controls in the Chinese Han population. Results: The proportions of smokers and drinkers in the OSCC patients were both significantly higher while compared with the control group. Moreover,the dominant model,the recessive model and the overdominant model results also showed marginal significant associations. In the stratified analysis,significant associations were found among the women,ages ≥ 60 years,smokers,and non-drinkers. Conclusion: rs2839698 polymorphism may be one susceptibility locus for OSCC in Chinese Han population. Long time tobacco and alcohol intake may increase the risk of OSCC.
