摘  要:目的:建立测定梅花鹿鹿茸和马鹿鹿茸药材中17种氨基酸含量的方法。方法:采用柱前衍生-高效液相色谱法,色谱柱为Aglient ZORABX SB-Aq C18(250 mm×4.6 mm,5μm),柱温为40℃,检测波长为254 nm,流动相A为0.1 mol/L醋酸钠缓冲液(pH6.5)-乙腈(93∶7),流动相B为乙腈-水(8∶2),流速为1 mL/min。结果:该方法线性关系良好(R2=0.9997),平均加样回收率为100.32%,RSD%为1.17%~4.01%。结论:鹿茸药材的质量受鹿的品种、自然环境、养殖技术等因素的影响。该方法适用于梅花鹿鹿茸和马鹿鹿茸中氨基酸的含量测定,可为质量控制提供保证。Objective:To establish a method for the determination of 17 amino acid contents in sika deer antler and red deer pilose antler.Methods:A pre-column derivatization-high performance liquid chromatography method was used.The column was Aglient ZORABX SB-Aq C18 (250mm×4.6mm,5μm).The column temperature was 40℃,the detection wavelength was 254nm,and the mobile phase A was 0.1mol/L.Sodium acetate buffer (pH 6.5)-acetonitrile (93:7),mobile phase B acetonitrile-water (8:2),flow rate 1mL/min.Results:The method has a good linear relationship (R^2=0.9997),with an average recovery of 100.32% and an RSD of 1.17% to 4.01%.Conclusion:The quality of velvet antler is influenced by deer species,natural environment,and breeding techniques.The method is applicable to the determination of amino acid content in sika deer pilose antler and red deer pilose antler to provide guarantee for the quality control of both.
