摘  要:目的探讨不同家栖鼠VKORC1基因片段大小是否存在差异,分析不同家栖鼠VKORC1基因启动子及第二外显子序列是否存在差异及与抗药性的关联。方法对捕自南通市3个区的家栖鼠提取尾组织DNA,采用sanger测序法对VKORC1基因启动子及第二外显子进行序列分析,通过SPSS16.0软件进行χ^2检验。结果黄胸鼠与褐家鼠的VKORC1基因片段大小存在差异。黄胸鼠VKORC1基因启动子序列共存在6处缺失突变和16个单核苷酸多态性(SNPs)。黄胸鼠第二外显子第90位氨基酸存在一个错义突变,3只抗药鼠均发生该错义突变,6只敏感鼠有3只发生了该错义突变。结论本研究发现VKORC1基因第二外显子第90位氨基酸的错义突变可能与鼠抗药性密切相关,有望为探讨家栖鼠发生抗药的分子机制奠定一定的基础。Objective To explore the difference in the length of VKORC1 gene fragment among different commensal rodents,and the sequence differences of the promoter and the second exon of VKORC1 gene among different commensal rodents and the association with anticoagulant resistance.Methods DNAs were extracted from tail tissues of commensal rodents captured from 3 districts of Nantong.Sanger sequencing method was adopted to analyze the sequences of the promoter and the second exon of VKORC1.SPSS 16.0 soft ware was used for χ^2 test.Results There was a difference in the length of VKORC1 gene fragment between Rattusflavipectus and Rattus norvegicus.Six deletion mutations and 16SNPs were found out in the promoter region of VKORC1 in Rattus flavipectus.The missense mutation in the 90th amino acid of the Rattus flavipectus was found in the three anticoagulant resistant rats,while the mutation occurred in three out of six susceptible rats.Conclusion This study revealed that the missense mutation of the 90th aminoacid in the second exon of VKORC1 gene might be associated with anticoagulant resistance,laying a theoretical foundation for the molecular mechanism study of the anticoagulant resistant of commensal rodents.
