摘 要:目的建立HPLC同时测定盐肤木提取物中没食子酸、苔黑酚葡萄糖苷、没食子酸乙酯和五没食子酰葡萄糖4种成分含量的方法。方法采用Hypersil-Gold Dim C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-0.1%甲酸,梯度洗脱,流速1.0 mL/min,柱温30℃,检测波长273 nm。结果没食子酸、苔黑酚葡萄糖苷、没食子酸乙酯、五没食子酰葡萄糖的进样量分别在0.025 7~0.514μg(r=1)、0.097 3~1.946μg(r=1)、0.040 1~0.801μg(r=1)、0.102 0~2.036μg(r=0.999 9)范围内呈良好的线性关系,平均加样回收率分别为99.86%(RSD=2.10%)、99.34%(RSD=1.91%)、98.80%(RSD=2.39%)、100.79%(RSD=2.04%),方法的精密度、稳定性、重复性均满足分析要求。结论该方法简单、快速、高效,结果准确,重复性好,可用于盐肤木提取物的质量控制。Objective To establish an HPLC method for simultaneous determination of gallic acid, ocinol glucoside, ethyl gallate and penta-O-galloyl-β-D-glucose from Rhus chinensis Mill extracts. Methods Hypersil-Gold Dim C18 column(4.6 mm × 250 mm, 5 μ m) was adopted. The mobile phase was composed of methanol and 0.1% formic acid solution with gradient elution at the flow rate of 1.0 mL/min. The column temperature was 30 ℃ and the detection wavelength was 273 nm. Results There was a good linear relationship between injection content and peak area when the gallic acid, ocinol glucoside, ethyl gallate and penta-O-galloyl-β-D-glucose were within the range of 0.025 7–0.514 μ g(r=1), 0.097 3–1.946 μ g(r=1), 0.040 1–0.801 μ g(r=1) and 0.102 0–2.036 μ g(r=0.999 9), respectively. The average recoveries were 99.86%(RSD=2.10%), 99.34%(RSD=1.91%), 98.80%(RSD=2.39%) and 100.79%(RSD=2.04%), respectively. The performance characteristics of precision, stability and reproducibility of the developed HPLC method fulfilled the analysis requirements. Conclusion The method is simple, rapid and efficient, with good reproducibility, and can be applicable for the quality control of Rhus chinensis Mill extracts.
