摘 要:目的探讨信号传导及转录激活因子5A(signal transducers and activators of transcription 5A,STAT5A)及NANOG基因mRNA在前列腺癌雄激素非依赖性转化过程中的作用。方法通过RT-PCR法比较LNCa P细胞与LNCa P-AI细胞中STAT5A及NANOG相关基因mRNA的差异表达,利用siRNA沉默STAT5A mRNA的表达检测LNCa P-AI细胞增殖情况的改变。结果 LNCa P-AI细胞较LNCa P细胞STAT5A、NANOG、OCT4基因mRNA均上调,差异倍数分别为(14. 60±9. 77)倍、(182. 33±98. 97)倍、(7. 44±1. 01)倍(P <0. 05);下调STAT5A后,SOX2、NANOG基因mRNA表达受抑制,实验组相对对照组表达量分别为(26. 60±13. 12)%、(15. 43±1. 89)%(P <0. 05)。CCK-8法检测沉默STAT5A后LNCa P-AI细胞,其生长受抑制,抑制率为(30. 24±6. 64)%,上述差异均有统计学意义(P <0. 05)。结论 STAT5A及NANOG在前列腺癌雄激素非依赖性转化过程中可能有重要作用,下调STAT5A mRNA的表达可以下调NANOG mRNA的表达,并且在一定程度上影响LNCa P-AI细胞的增殖。Objective To investigate the role of signal transducers and activators of transcription 5A(STATSA)and NANOG mRNA in androgen-independent transformation of prostate cancer.Methods The mRNA expressions of STATSA and NANOG in LNCaP cells and LNCaP-AI cells were compared by RT-PCR.The expression of STATSA mRNA was detected by siRNA to observe the proliferation changes of LNCaP-AI cells.Results The mRNA levels of STATSA,NANOG and OCT4 in LNCaP-AI cells were up-regulated by (14.60±9.77)times,(182.33±98.97)times and (7.44±1.01)times (P<0.05).After down-regulating STATSA,the mRNA expressions of SOX2and NANOG were inhibited.The expressions of them in the experimental group to that in the control group were (26.60±13.12)%and (15.43±1.89)%(P <0.05).The growth of LNCaP-AI cells was tested by CCK-8assay,and the inhibition rate of the growth was (30.24±6.64)%,and the above differences were statistically significant (P <0.05).Conclusion STATSA and NANOG genes may play an important role in the androgen-independent transformation of prostate cancer.Down-regulation of STATSA mRNA may down-regulate the expression of NANOG mRNA and may affect the proliferation of LNCaP-AI cells to a certain extent.
