摘 要:为了解2015~2017年湖北地区地方品种鸡中J亚群禽白血病病毒(Subgroup J avi-an leukosis vrius,ALV-J)流行情况及流行株的分子特征,从湖北省地方品种鸡中分离到10株ALV-J,对其gp85基因进行PCR扩增、克隆、序列测定及分析。结果显示:10个分离毒株的gp85基因片段大小均为924 bp,与预期一致;10个分离毒株之间核苷酸和氨基酸序列相似性分别为91.4%~100%、86.4%~100%,其中9株与原型毒株HPRS103核苷酸相似性为96.8%~98.9%,并且在一个分支上面。结果表明:湖北省部分地方品种鸡不同程度感染ALV-J,分离毒株的变异主要围绕着原型株HPRS103改变,应加强地方品种鸡ALV的净化工作。To investigate the epidemiology of subgroup J avian leukosis vrius(ALV-J) in 2015 to 2017 in Hubei area,10 strain viruses were isolated and detected from local chicken breeds. The results of PCR showed that gp85 genes ofthe ten strains were all 924 bp. Sequence analysis revealed that the strains shared 91.3% to 100% nucleotide similarityand 86.4% to 100% similarity at amino acid level. Morever,the nine strains were 96.8% to 98.9% nucleotide similarwith the prototype strain HPRS103. JM6194 strain had the longest genetic distance to other samples. In conclusion,thelocal chicken breedsn in Hubei area were infected with ALV-J to varying degrees. The variation of the isolated strainswas mainly changed around the prototype strain HPRS103, and the purification of ALV-J in the local chicken breedsshould be strengthened.
