摘　　要：目的探讨转化生长因子β(transforming growth factor-β,TGF-β)在弥漫大B细胞淋巴瘤中表达,确定TGF-β诱导体内调节性T淋巴细胞(Treg细胞)促进淋巴瘤生长的作用。方法分别采用RT-PCR和Western Blotting方法检测TGF-β在正常B细胞及弥漫性大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)细胞株OCI-Ly1、OCI-Ly8、OCI-Ly10中的表达,免疫组化方法检测TGF-β蛋白在40例DLBCL患者淋巴结及健康人群淋巴结组织石蜡标本中的表达,流式细胞仪检测健康人群及DLBCL患者外周血淋巴细胞悬液中CD4^+CD25+Foxp3^+Treg细胞比值。结果 TGF-β在DLBCL细胞株OCI-Ly1、OCI-Ly8、OCILy10中较正常B细胞显著增高(P<0.05),TGF-β蛋白在DLBCL组织中较正常淋巴组织显著增高(P<0.05),DLBCL患者CD4^+CD25^+Foxp3^+Treg细胞比值高于健康人群(P<0.05)。结论 TGF-β在DLBCL细胞系及组织中均呈高表达。TGF-β可能诱导人体内的淋巴细胞成为Treg细胞,从而促进DLBCL的发生。Objective To investigate the expression of TGF-β in diffuse large B cell lymphoma,and the effect of TGF-β on the induction of regulatory cells (Treg cells)and the growth of lymphoma.Method Detected the expression of TGF-β in normal B cells and OCI-Ly1, OCI-Ly8,OCI-Ly 10 cell lines of DLBCL by RT-PCR and Western Blotting.Immunohistochemistry was used to detect the expression of TGF-β protein in 40 paraffin specimens of DLBCL lymph nodes and normal lymph nodes.Flow cytometry was used to detect the number of CD4^+CD25^+Foxp3^+Treg cells in lymphocytes of healthy people and DLBCL patients.Result TGF-β was significantly higher than normal B cells in DLCCL cell lines OCI-Ly1,OCI-Ly8,and OCI-Ly10(P<0.05),and TGF-β protein was significantly higher in DLBCL than normal lymphoid tissues (P<0.05).The number of CD4^+ CD25^+Foxp3^+ Treg cells in DLBCL patients was higher than that in normal subjects (P<0.05).Conclusion TGF-β was highly expressed in DLBCL cell lines and tissues.TGF-β may induce human lymphocytes to become Treg cells and promote the occurrence of DLBCL.
