金雀异黄素对肺癌细胞生长、侵袭和转移的影响

摘　　要：目的探讨金雀异黄素对人肺癌细胞生长、侵袭和转移的影响及其机制。方法金雀异黄素处理体外培养的人肺癌A549和H358细胞,采用MTT法检测细胞活力,集落形成实验观察细胞生长,流式细胞术分析细胞凋亡,细胞迁移与侵袭实验观察对肺癌细胞迁移和侵袭的影响。在裸鼠体内注入绿荧光素酶标记的肺癌A549细胞,观察金雀异黄素对肺癌细胞体内生长和转移的影响。采用qRT-PCR和Western blot分析金雀异黄素对肺癌细胞转移相关mRNA和蛋白表达的影响。结果金雀异黄素可以抑制肺癌A549和H358细胞的生长。金雀异黄素能引起肺癌细胞凋亡增加(P<0.01)。金雀异黄素可以抑制体外和体内肺癌细胞的迁移和侵袭(P<0.01)。金雀异黄素能下调MMP-9和血管内皮生长因子受体3(VEGFR3)mRNA和蛋白的表达。结论金雀异黄素能抑制肺癌细胞生长、侵袭和转移;其机制可能与下调MMP-9和VEGFR3的表达有关。Objective To explore the effect and underlying mechanism of genistein on the growth,invasion and metastasis of human lung cancer cells.Methods Lung cancer cell lines A549 and H358 were cultured and treated by genistein in vitro.Cell viability was analyzed using MTT assay. Colony formation assay was used to observe the growth of lung cancer cells.Flow eytometry was performed to determine the apoptosis of cells.Cell migration and invasion assay was performed to observe the effects of genistein on the migration and invasion of lung cancer cells.The effect of genistein on the growth and migration of lung cancer cells in vivo was observed by injecting the green luciferase-labeled A549 lung cancer cells into nude rats.qRT-PCR and Western blot were used to analyze the effect of genistein on the expressions of metastasis-related mRNA and protein in lung cancer cells.Results Genistein inhibited the growth of A549 and H358 cells.Genistein could induce A549 and H358 cells apoptosis (P<0.01).Genistein inhibited the growth and metastasis of lung cancer cells both in vitro and in vivo (P<0.01).Moreover,genistein could downregulate the mRNA and protein expressions of MMP-9 and vascular endothelial growth factor receptor 3(VEGFR3) showed by qRT-PCR and Western blot,respectively.Conclusion Genistein is able to inhibit the growth,invasion and metastasis of human lung cancer cells,which may be related to the downregulation of MMP-9 and VEGFR3 expressions.
