肝素和无水乙醇溶液体外抑制肝癌细胞的效应

摘　　要：目的探讨不同浓度肝素和无水乙醇溶液对体外肝癌细胞的抑制效应。方法以PBS溶液处理作为对照,用50U/ml肝素溶液作用肝癌细胞(huh7细胞)和正常肝细胞(L02细胞)3h,检测细胞存活数。以PBS溶液处理作为对照,分别采用浓度为10、50和250U/ml的肝素溶液或75%乙醇和无水乙醇作用huh7细胞3h,检测huh7细胞存活数。结果与PBS溶液组比较,采用50U/ml的肝素溶液处理后,huh7细胞存活数减少(P<0.05),而L02细胞存活数与之相仿(P>0.05)。与PBS溶液组比较,采用10U/ml的肝素溶液处理后,huh7细胞存活数与之相仿(P>0.05),但50U/ml和250U/ml肝素组huh7细胞存活数减少(P<0.05),75%乙醇和无水乙醇组的huh7细胞存活数减少更为明显(P<0.01)。结论采用浓度为50U/ml和250U/ml的肝素溶液处理能明显减少体外肝癌细胞的存活。Objective To test the inhibitory effect of heparin and anhydrous ethanol solution on hepatocellular carcinoma cells in vitro.Methods Taking the treatment with PBS solution as the control,the hepatocellular carcinoma cell line huh7 and normal hepatic cells(L02)were treated with50 U/ml heparin for 3 hours and the survival numbers of the cells were detected.Taking PBS solution treatment as the control,the huh7 cells were treated with different concentrations of heparin(10 U/ml,50 U/ml and 250 U/ml)and anhydrous ethanol solution(75% and 100%)for 3 hours and the survival numbers of the huh7 cells were detected.Results Compared to the group treated with PBS solution,the survival number of huh7 cells was less in the group treated with 50 U/ml heparin(P<0.05),but that of L02 cells was similar(P >0.05).Compared to the group treated with PBS solution,the survival number of huh7 cells in the group treated with 10 U/ml heparin was similar(P >0.05),which was significantly less in the groups treated with 50 U/ml and 250 U/ml heparin solutions(P<0.05)or treated with 75% and 100% anhydrous ethanol solutions(P<0.01).Conclusion The survival of hepatocellular carcinoma cells is remarkably reduced when they are treated with 50 U/ml and 250 U/ml heparin solutions.
