摘 要:目的探讨七氟醚对乳腺癌细胞NF-κB活性及基质金属蛋白酶(MMPs)表达的影响,为乳腺癌治疗提供新的参考数据。方法体外培养乳腺癌细胞AU565,分别用1. 7%、3. 4%、5. 1%七氟醚作用4 h,分别设为低剂量组、中剂量组、高剂量组,将未经七氟醚作用培养的细胞设为对照组。利用MTT检测各组细胞增殖情况,实时荧光定量分别检测MMP-2、MMP-9 mRNA相对表达量,蛋白免疫印迹检测IKKβSer180、NF-κB p65、MMP-2、MMP-9蛋白表达情况,EMSA检测NF-κB活性。蛋白免疫印迹检测IL-1β预处理细胞经3. 4%七氟醚作用后,IKKβSer180、NF-κB p65、MMP-2、MMP-9蛋白表达变化。结果与对照组相比,低剂量组、中剂量组、高剂量组细胞增殖率均显著下降,差异有统计学意义(P<0. 05);与对照组相比,低剂量组、中剂量组、高剂量组中MMP-2、MMP-9 mRNA相对表达量显著下调,差异有统计学意义(P<0. 05),IKKβSer180、NF-κB p65、MMP-2、MMP-9蛋白表达量显著下降,差异有统计学意义(P<0. 05),NF-κB活性显著降低,差异有统计学意义(P<0. 05)。IL-1β预处理后,3. 4%七氟醚对乳腺癌细胞中MMP-2、MMP-9蛋白表达量的抑制作用显著减弱。结论七氟醚是一种潜在的治疗乳腺癌的分子靶向药物,可通过抑制NF-κB活性来降低MMPs表达。Objective To explore the effect of sevoflurane on breast cancer NF-κB cell activity and expressions of MMP2 and MMP9,provide a new reference basis for treatment of breast cancer.Methods Breast cancer cell line AU565 was cultured in vitro,and all the cells were treated by 1.7%,3.4%,and 5.1% sevoflurane for 4 hours,respectively(low-dose group,medium-dose group,high-dose group),and the cells cultured without sevoflurane were enrolled as control group.Cell proliferation was detected by MTT,the relative expression levels of MMP-2 and MMP-9 mRNA were detected by real-time quantitative fluorescence.The expression levels of IKKβ Ser180,NF-κB p65,MMP-2,and MMP-9 protein were detected by Western blotting.NF-κB activity was detected by EMSA.The changes of IKKβSer180,NF-κB p65,MMP-2,MMP-9 protein expressions were detected by Western blotting after pretreated by IL-1β and treated by sevoflurane.Results Compared with control group,cell proliferation rates in low-dose group,medium-dose group,and high-dose group decreased significantly(P<0.05).Compared with control group,the relative expression levels of MMP-2 and MMP-9 mRNA in low-dose group,medium-dose group,and high-dose group down-regulated significantly(P<0.05).The expression levels of IKKβ Ser180,NF-κB p65,MMP-2 and MMP-9 proteins decreased significantly(P<0.05),and NF-κB activity decreased significantly(P<0.05).After pretreated by IL-1β,3.4% sevoflurane significantly reduced the inhibitory effect on expression levels of MMP-2 and MMP-9 proteins in breast cancer cells.Conclusion Sevoflurane is a potential molecular targeted drug in treatment of breast cancer,and it can reduce MMPs expressions by inhibiting the activity of NF-κB.
