摘 要:目的探讨远隔缺血后处理(remote ischemic postconditioning,RIPC)对未行血管再通的急性缺血性脑卒中小鼠是否具有脑保护作用。方法线栓法致小鼠MCAO制备大脑局部缺血模型,并进行神经功能缺损控制再灌注损伤评分(Bederson评分),将制成的模型鼠随机分为两组:远隔缺血同处理组(RIPC组,n=15):插入线栓30 min后给以小鼠左下肢缠绕止血带,4 min缺血处理,4 min再灌注,3个循环。假缺血处理组:(Sham组,n=11),给以左下肢缠绕止血带,但不扎紧,给以假处理。24 h后对两组小鼠进行Bederson评分,过量麻醉处死后取脑,行TTC染色,测定梗死体积。结果①脑梗死体积测定:RIPC组小鼠脑梗死体积小于Sham组小鼠,差异有统计学意义(P<0.05);②Bederson评分:RIPC组小鼠与Sham组小鼠功能评分相近,差异无统计学意义(P>0.05)。结论远隔缺血后处理在没有血流再灌情况下能够减少梗死体积。Objective To investigate the neuroprotective roles of remote ischemic preconditioning(RIPC) in permanent middle cerebral artery occlusion(pMCAO) model mice.Methods The pMCAO mice were randomly divided into two groups:RIPC group(n=15) and sham group(n=11).In RIPC group:mice’s left leg was wrapped by tourniquet after inserting the string for 30 minutes,ischemia treatment for 4 minutes,repercussion for 4 minutes,3 circulations.In sham group:mice’s left leg was loosely wrapped by tourniquet.The Bederson scores were measured after 24 h of MCAO.The infarct volume was recorded using 2,3,5-triphenyltetrazolium chloride(TTC) staining.Results ①The cerebral infarct volume of RIPC mice was significantly decreased compared with the sham group(P<0.05);②The RIPC mice had no significant difference in Bederson scores compared with the sham control mice(P>0.05).Conclusion RIPC could reduce the cerebral infarct volume following ischemia.
