摘  要:目的通过对人胰腺癌细胞系Bx PC-3诱导吉西他滨耐药细胞黏附性质变化,初步探讨其耐药的分子生物学机制。方法 0. 2μmol/L吉西他滨培养基诱导2个月获得Bx PC-3细胞系的耐药细胞株Bx PC-3-GEM,观察细胞的生长状态,通过ELISA、Western blot等方法测定细胞产生的可溶性细胞间黏附因子-1(s ICAM-1)和E-钙黏连蛋白(E-cadherin),应用细胞集落形成以及黏附实验判断细胞的黏附特点。结果与亲代细胞株比较,耐药细胞株Bx PC-3-GEM细胞大小无明显变化,仍呈现聚集性生长,但胞浆内颗粒较大,细胞核明显、异形细胞核增多。体外增殖能力减弱,分泌的s ICAM-1减少。结论通过吉西他滨诱导获得的胰腺癌Bx PC-3-GEM细胞黏附率下降,影响了体内肿瘤细胞的转移。Objective To explore the change of cell adhesion of gemcitabine-induced human pancreatic cancer cell line BxPC-3 and discuss the molecular mechanism of drug resistance.Methods The cell line BxPC-3-GEM was induced by BxPC-3 cultured in the medium with0. 2 μmol/L gemcitabine for 2 months,and the growth of cells was observed. The soluble intercellular adhesion 1( s ICAM-1) and E-cadherin detected by ELISA or Western blot were used to determine the cell adhesion characteristics by cell colony formation and adhesion assay. Results There was no significant change in the size of BxPC-3-GEM cells compared with the parental cell lines,still showed aggregated growth,but the intracytoplasmic granules were larger,the nuclei were obvious and the number of abnormal nuclei increased. At the same time,the in vitro proliferation decreased and the secretion of s ICAM-1 decreased. Conclusion The cell adhesion rate of pancreatic cancer BxPC3-3-GEM cells induced by gemcitabine decreases,which affects the metastasis of tumor cells in vivo.
