摘 要:目的 采用3.0T MR动态观察C57BL/6小鼠前列腺癌皮下移植瘤的生物学特征。 方法 采用前列腺癌RM-1细胞悬液接种于50只C57BL/6小鼠的背部皮下,待触及皮下移植瘤时,每5 d采用游标卡尺测量移植瘤的长径和短径,计算移植瘤体积;MR在第15、20、25及35 d对移植瘤进行测量和观察。移植术后第35天处死小鼠,并绘制移植瘤生长曲线。处死小鼠后观察移植瘤大小、形态,有无局部侵袭及淋巴、脏器转移,并行病理学检查。采用游标卡尺体外测量移植瘤体积并与MR测量结果进行对照,比较其差异。 结果 移植瘤多呈圆形或椭圆形,平均成瘤潜伏期为(13.41±1.37)d,移植瘤成功率为90%,瘤体倍增时间为10 d左右。MR检查移植瘤直径≤1.5 cm时形态规则,信号尚均匀;当移植瘤直径约2.0 cm时其内部见不均匀混杂信号,符合坏死表现。游标卡尺体外测量、MR测量2种方法所得移植瘤体积结果的比较无统计学差异(P>0.05)。第35天MR测量移植瘤体积与病理测量结果的相关性(r=0.971,P<0.01)较游标卡尺体外测量与病理测量结果的相关性更高 (r=0.906,P<0.05)。 结论 3.0T MR能无创、清晰地显示小鼠前列腺癌移植瘤的生长变化情况,能够提供更多的信息来评价其生物学特征。Objective To observe the biological characteristics of prostate cancer subcutaneous xenograft in C57BL/6 mice by 3.0T MR.Methods The prostate cancer cell line RM-1 cells were inoculated into the back of 50 C57BL/6 mice,when the subcutaneous xenograft were to be touched,the long diameter and the short diameter of the xenograft were measured with a vernier caliper every 5d, and the volume of the xenograft was calculated.The xenografts were measured and observed on days 15,20,25 and 35.The mice were sacrificed on the 35th day after transplantation and the growth curve of the xenograft was drawn.After the mice were sacrificed,the size and shape of the xenografts,with or without local invasion and lymph node metastasis were observed,and pathological examination was performed.The volume of xenografts were measured by vernier caliper and compared with the results of MR.Results The xenografts were mostly round or oval,with an average tumor-forming latency of (13.41±1.37)d,the success rate of the xenograft was 90% and the tumor doubling time was about 10 d.MR examination showed that the morphology of the xenografts were regular and the signal were uniform when the diameter was ≤1.5 cm.When the xenografts were about 2.0 cm in diameter,they showed heterogeneous mixed signals,which was consistent with necrotic performance.There was no statistical difference in the volume of the xenografts obtained by the two methods of vernier caliper measurement and MR measurement (P>0.05).On day 35,the correlation between MR measurement and pathological measurement of xenograft volume was higher (r=0.971,P<0.01)than that of vernier calipers in vitro and pathological measurements (r=0.906,P<0.05).Conclusion 3.0T MR can noninvasively and clearly show the growth and change of prostate cancer xenograft in mice,it can provide more information to evaluate its biological characteristics.
