摘　　要：目的探讨硫酸右旋糖酐(DS)通过影响Twist抑制人胃癌细胞(BGC-823)上皮间质转化(EMT)的作用及机制。方法培养人胃癌细胞(BGC-823),对照组和实验组分别用PBS和DS进行处理,低氧培养2、8、12、24 h,用细胞免疫荧光进行染色和Western blot方法检测DS对EMT相关因子(Twist、E-cad、N-cad、)蛋白表达水平的影响。结果细胞免疫荧光染色和Western blot实验结果显示,实验组比对照组,Twist、N-cad的表达均表现出不同程度的减少,E-cad的表达水平表现出不同程度的增加,差异有统计学意义。结论 DS可能通过影响Twist抑制人胃癌BGC-823细胞的EMT。Objective To investigate the inhibitory effect of Dextran Sulfate on Epithelial-Interstitial Transformation of Human gastric Cancer cells via Twist and its Mechanism. Methods Human gastric cancer cells( BGC-823) were cultured and treated with PBS and DS in the control group and experimental group,respectively. The cells were cultured in hypoxia for 2h,8h,12h,24h. The effect of DS on emt-related factor protein( Twist、E-cad、N-cad) expression was detected by cell immunofluorescence staining and Western blot.Result The results of cell immunofluorescence staining and Western blot showed that the expressions of Twist and N-cad in the experimental group were decreased to different degrees,and the expression levels of E-cad were increased to different degrees,with statistically significant. Conclusion DS can inhibit bgc-823 cell EMT in human gastric cancer by influencing Twist
