摘 要:目的探讨Notch蛋白和Snail蛋白表达水平对胃癌细胞迁移能力的影响,为治疗胃癌提供参考。方法选取2014年4月至2016年4月本院实施胃癌切除术的62例患者为研究对象,术后采集患者胃癌及癌旁组织,用超微量分光度计对mRNA进行测定,用BCA蛋白测量法测定Notch和Snail蛋白含量。观察记录患者的姓名、年龄、身高、体重、癌症临床分期、肿瘤细胞浸润深度、分化程度等指标。结果 Notch1、Notch2、Notch3和Snail在胃癌组织中的表达率明显高于癌旁组织,差异有统计学意义(P<0.05)。患者癌症临床分期越晚、肿瘤细胞浸润越深,Notch1蛋白表达水平和阳性表达率越高,差异有统计学意义(P<0.05);Notch2蛋白表达水平与阳性表达率与患者临床特征无关;Notch3蛋白表达水平和阳性表达率与患者癌症分化程度、性别、癌症临床分期、肿瘤细胞浸润深度和淋巴结转移无关,差异无统计学意义(P>0.05),与患者年龄相关,差异有统计学意义(P<0.05)。Notch和Snail蛋白在胃癌中表达呈正相关(r=0.462,P=0.002)。结论 Notch和Snail蛋白表达水平与胃癌患者癌症临床分期和肿瘤浸润深度等临床特征密切相关,在一定程度上能准确反映患者肿瘤状态,为胃癌的治疗提供参考。Objective The effects of Notch protein expression on Snail and the migration ability of gastric cancer cells in vivo were analyzed, and provide a theoretical basis for the treatment of gastric cancer. Methods A total of 62 patients who underwent curative resection of gastric cancer from April 2014 to April 2016 were selected as subjects. Patients with gastric cancer and paracancerous tissues were collected after surgery, and mRNA was measured with an ultra-micrometer spectrophotometer. The Notch and Snail protein content was determined using the BCA protein assay. Patient’s name, age,height, weight, clinical stage of cancer, deep infiltration of tumor cells, and the degree of differentiation were observed and recorded. Results The expression rates of Notch1, Notch2, Notch3 and Snail in gastric cancer tissue were significantly higher than those in paracancerous tissues(P<0.05). The later the clinical stage of cancer, the deeper the tumor cell infiltration, the higher the expression level and positive expression rate of Notch1 protein, the difference was statistically significant(P<0.05). The expression level of Notch2 protein and the positive expression rate were not related to the clinical features of cancer in patients. The expression level of Notch3 protein and the positive expression rate were not related to the degree of cancer differentiation, gender, clinical stage, tumor cell infiltration and lymph node metastasis, the difference was not statistically significant(P>0.05), however they were related to the age of the patients;the difference was statistically significant(P<0.05). There was a positive correlation between Notch and Snail protein expression in gastric cancer(r=0.462,P=0.002). Conclusions The expression levels of Notch and Snail protein were closely related to the clinicopathological features such as clinical stage and depth of tumor invasion in gastric cancer patients.
