摘 要:目的探讨跨膜蛋白158(TMEM158)在乳腺癌中的表达及与临床特征及预后的相关性。方法选取2013年4月至2015年4月于本院诊治的105例乳腺癌患者作为研究对象,采用实时荧光定量聚合酶链反应(qRT-PCR)法和免疫组织化学法(IHC)检测乳腺癌组织和癌旁组织中TMEM158 mRNA和蛋白的表达差异,并分析其与临床特征及预后的关系。结果乳腺癌组织中TMEM158 mRNA表达水平(7.05±1.13)和TMEM158蛋白阳性率(68.57%)明显高于癌旁组织[(4.25±0.75)、(9.52%)],差异有统计学意义(P<0.05);TMEM158蛋白阳性表达者和阴性表达者在肿瘤大小、淋巴转移、临床分期、分子分型、孕酮受体(PR)、雌激素受体(ER)和Ki-67上差异均有统计学意义(P<0.05);TMEM158蛋白阳性者无瘤生存时间明显低于TMEM158蛋白阴性者(χ~2=5.242,P=0.022);经Cox回归分析得知,肿瘤大小、临床分期、淋巴转移和TMEM158蛋白是影响乳腺癌预后的危险因素(P<0.05)。结论TMEM158在乳腺癌中高表达,与不良预后密切相关,是影响乳腺癌预后的危险因素。Objective To study the expression of transmembrane protein 158(TMEM158) in breast cancer and its correlation with clinical characteristics and prognosis. Methods 105 cases of breast cancer in our hospital from April 2013 to April 2015 were selected as subjects. The expression of TMEM158 in breast cancer tissues and adjacent normal tissues was detected by the qRT-PCR and IHC;the relationship between the expression and the clinical characteristics and prognosis was analyzed. Results The expression of TMEM158 mRNA(7.05 ± 1.13) and TMEM158 protein positive rate(68.57%) in breast cancer tissues were significantly higher than those of normal tissue[(4.25±0.75),(9.52%)],and the difference was statistically significant(P<0.05). There had a great difference between tumor size, lymph node metastasis,clinical stage, molecular typing, progesterone receptor(PR), estrogen receptor(ER) and Ki-67 in TMEM 158-positive and TMEM 158-negative cases(P<0.05). The tumor free survival time of TMEM158 positive patients was significantly lower than that of TMEM 158 negative patients(χ^2 =5.242, P=0.022). The COX regression analysis showed that tumor size, clinical stage, lymph node metastasis and TMEM 158 protein were the risk factors for the prognosis of breast cancer(P<0.05).Conclusion The expression level of TMEM 158 was high in breast cancer, and was closely related to poor prognosis, and it could be a key independent risk factor affecting the prognosis of breast cancer.
