摘 要:目的:探究穿心莲内酯及类似物对Chang Liver细胞活力的影响和机制。方法:CCK-8法检测穿心莲内酯、新穿心莲内酯、脱水穿心莲内酯、脱水穿心莲内酯琥珀酸半酯对Chang Liver细胞活力的影响;Q-PCR和Western blot法分别检测药物不同浓度对Chang Liver细胞中CDK1、CDK2、CDK4、CDK6 mRNA和蛋白表达的影响。结果:穿心莲内酯、脱水穿心莲内酯和脱水穿心莲内酯琥珀酸半酯呈浓度依赖性地抑制细胞增殖,新穿心莲内酯对细胞增殖无影响;脱水穿心莲内酯、脱水穿心莲内酯琥珀酸半酯在药物达到一定剂量时均可抑制CDK4与CDK6 mRNA水平的表达;穿心莲内酯、脱水穿心莲内酯与脱水穿心莲内酯琥珀酸半酯在100μmol/L时,均可抑制CDK4与CDK6蛋白水平的表达;新穿心莲内酯对CDKs的抑制不明显。结论:穿心莲内酯、脱水穿心莲内酯、脱水穿心莲内酯琥珀酸半酯对细胞增殖的抑制作用与对CDK4、CDK6的调节密切相关。Objective: To study the effects and mechanism of Andrographolide and its analogues on the viability of Chang Liver cells. Methods: CCK-8 assay was performed to determine the proliferation of Chang Liver cells after treatment with Andrographolide, Neoandrographolide, Dehydroandrograph, Dehydroandrographolide succinate. Q-PCR and Western blot were respectively performed to examine the effects of different concentrations of drugs(10 and 100 μmol/L) on CDK1, CDK2, CDK4, CDK6 mRNA and protein expression in Chang Liver cells.Results: Andrographolide, Dehydroandrograph and Dehydroandrographolide succinate displayed significantly inhibitory effects on hepatic growth at the concentration of 40 ~80 μM, however, Neoandrographolide had no inhibitory effect even at 100 μmol/L. Dehydroandrograph and Dehydroandrographolide succinate could decrease the level of CDK4 and CDK6 mRNA. Andrographolide, Dehydroandrograph and Dehydroandrographolide succinate could reduce the protein expressions of CDK4 and CDK6. However, Neoandrographolide had not show inhibitory effects on CDKs. Conclusion: Andrographolide, Dehydroandrograph and Dehydroandrographolide succinate can inhibit proliferation of Chang Liver cells, and the effects are closely related to regulation of CDK4 and CDK6.
