摘 要:目的研究姜黄素与5-FU联用对人结肠癌细胞自噬及对YAP的影响。方法姜黄素、5-FU单独应用及两者联用作用于人结肠癌HCT116细胞和过表达YAP的结肠癌稳定HCT116细胞系24 h,MTT法检测细胞增殖情况,Western blot检测自噬标志物LC3-Ⅱ的变化,并检测癌基因YAP表达变化。结果 MTT法检测结果显示,2种药物联合应用后,肿瘤细胞的增殖抑制率明显高于单用药组(P<0.01)。姜黄素组、5-FU组和药物联用组HCT116细胞的YAP蛋白表达水平下降,LC3-Ⅱ蛋白的表达水平增高。结论姜黄素和5-FU联用会下调YAP的表达,并诱导结肠癌细胞自噬的表达,YAP或许能成为结肠癌治疗的一个新的靶点。This study was designed to analyze the effects of curcumin and 5-FU on the autophagy and YAP of colon cancer cells. The inhibitory effects of curcumin and/or 5-FU on colon cancer cell line HCT116 were investigated using MTT methods;the changes of LC3-Ⅱ and YAP expression after the treatment was also evaluated by Western blotting. Data showed that proliferation of HCT116 cells was significantly inhibited by curcumin and 5-FU in combination. In addition, the expression level of YAP protein was reduced after the use of the two agents in combination, and the level expression of LC3-Ⅱ protein was increased after the same treatment. Our results suggest that curcumin and 5-FU in combination can down-regulate YAP expression, which may has great promise for treatment of colon cancer and might be a potential diagnostic marker for colon cancer.
