摘 要:肉桂酸4-羟基化酶(cinnamate 4-hydroxylase, C4H)是植物苯丙烷代谢途径中的关键酶,能够影响类黄酮次生代谢物的生物合成。本研究根据海南龙血树转录组数据,利用RT-PCR技术在海南龙血树中克隆了一个C4H基因DcC4H,该基因包含一个1 518 bp的开放阅读框,编码505个氨基酸,蛋白质分子量57.82 kD。生物信息学分析显示,DcC4H属于细胞色素氧化酶P450 Superfamily,具有Cytochrome P450保守位点;蛋白序列比对显示DcC4H与其它植物C4H同源性高,保守性强。表达结果显示,DcC4H在花中的表达显著高于根、叶、果和茎;脱落酸(ABA)、6-苄氨基腺嘌呤(6-BA)、油菜素内酯(BR)、茉莉酸(JA)、紫外线(UVB)和蔗糖处理下,DcC4H的表达量均上调,其中UVB处理表达上调最显著。本研究将为进一步研究DcC4H在海南龙血树类黄酮生物合成的功能提供理论依据。Cinnamic acid 4-hydroxylase is a key enzyme in the phenylpropane metabolic pathway. C4 H affect the biosynthesis of flavonoid secondary metabolites. Based on the transcriptional data from Dracaena cambodiana, a C4 H gene designated as DcC4 H was cloned by RT-PCR technology. DcC4 H contained a 1 518 bp open reading frame encoding 505 amino acids with a predicted molecular mass of 57.82 kD. Bioinformatics analysis showed that DcC4 H belonged to P450 superfamily and had the conserved site of cytochrome P450. DcC4 H was highly homologous to other plant’s C4 H. DcC4 H was significantly expression in flower, followed by root, leaf, fruit and stem.The transcription level of DcC4 H increased after induced by ABA, BR, 6-BA, JA, UVB and sucrose. This study will lay the foundation for further study the function of DcC4 H in Dracaena cambodiana flavonoid biosynthesis.
