摘　　要：神经系统疾病是神经系统由于感染、免疫、血管病、代谢障碍等引起的一类综合性疾病。随着我国人口老龄化的到来,神经系统疾病已成为致死和致残的主要原因之一。神经系统疾病的发病机制非常复杂,研究发现许多基因或信号通路的异常和该疾病的发生和发展密切相关。其中,程序性死亡受体-1(programmed death-1,PD-1)信号通路对人体免疫系统起负性调节作用,有利于维持免疫耐受和免疫自稳。因此PD-1及其配体的基因变异与神经系统免疫性疾病、退行性病变及脑血管病的发生关系密切,特别是在一些因感染和免疫异常引起的神经系统疾病的发生过程中,作用更加重要。因此,总结PD-1信号通路及相关基因在神经系统疾病中的作用,揭示PD-1信号通路对神经系统疾病发生、发展的影响,将为疾病的诊断、治疗和预后提供思路和帮助。Nervous system disease is a comprehensive disease of nervous system caused by infection,immunity,vascular diseases and metabolic disorders,etc.With the coming of aging population in China,nervous system disease has become one of the main causes of death and disability.The pathogenesis of nervous system disease is very complicated.Studies have shown that the abnormalities of many genes or signaling pathways have close relationship with the occurrence and development of the disease.The programmed death-1(PD-1) signaling pathway plays a negative role in regulating the immune system,which contributes to maintain immune tolerance and homeostasis.Therefore,the genetic variation of PD-1 and its ligands are closely related to the occurrence of immune diseases,degenerative diseases,and cerebrovascular diseases of the nervous system,especially in the process of some nervous system diseases caused by infection and immune abnormality.Here we summarize the role of PD-1 signaling pathway and its related genes in nervous system diseases.At the same time,we reveal the influence of PD-1 signaling pathway on the occurrence and development of nervous system diseases.
