摘 要:目的近年来非编码RNA在肿瘤中的作用逐渐被揭示,尤其是微小RNA(microRNA;miRNA)及长链非编码RNA(1ong noncoding RNAs,lncRNAs)与肿瘤的发生发展密切相关。本研究探讨微小RNA155-5p(microRNA-155-5p,miR-155-5p)及其宿主长链非编码RNA MIR155HG在喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)组织中的表达及临床意义,为探讨喉癌的发生机制提供新的思路。方法收集2016-09-01-2017-12-30河北医科大学耳鼻咽喉头颈外科生物样本库40例LSCC及癌旁正常组织标本,应用实时荧光定量反转录PCR技术(RT-qPCR)检测LSCC及癌旁正常组织中miR-155-5p及MIR155HG的表达水平,并分析其表达与LSCC患者临床病理参数间的关系,同时分析miR-155-5p及MIR155HG表达之间的相关性。结果 LSCC组织中miR-155-5p的相对表达量为3.158±1.049,显著高于癌旁正常组织1.058±0.636,差异有统计学意义,t=4.001,P<0.05;喉鳞癌组织中MIR155HG的相对表达量为3.865±1.012,显著高于癌旁正常组织1.686±0.456,差异有统计学意义,t=2.818,P<0.05;LSCC组织中miR-155-5p与MIR155HG的表达与年龄、吸烟和饮酒无关,差异无统计学意义,P>0.05;与TNM分期、肿瘤分化、淋巴结转移相关,差异有统计学意义,P<0.05;MIR155HG与miR-155-5p的表达呈正相关,r=0.654 3,P<0.05。结论 miR-155-5p及其宿主基因lncRNA MIR155HG表达上调与喉癌的发生和侵袭转移密切有关,而且MIR155HG与miR-155-5p的表达具有一致性,二者有可能成为新的喉癌靶向治疗肿瘤标志物。OBJECTIVE The role of non-coding RNA,especially miRNA and lncRNA,which are closely related to the development of tumor,has been gradually revealed in tumor in recent years.This study was to explore the expression and clinical significance of microRNA-155-5p and its host gene long noncoding RNA MIR155HG in larynx squamous cell carcinoma(LSCC)and provide new ideas for the occurrence of laryngeal carcinoma.METHODS Forty cases were collected from otolaryngology head and neck surgery biobank of Hebei Medical University from 2016-09-01 to 2017-12-30.The expression of microRNA-155-5p and MIR155HG in LSCC tissues and corresponding adjacent normal tissues were detected by real-time quantitative PCR.The relationship between the expression of microRNA-155-5p/MIR155HG and the clinical pathological characteristics in LSCC as well as the correlation between the expression of miR-155-5p and MIR155HG were analyzed.RESULTS The relative expression of miR-155-5p in LSCC tissues (3.158±1.049)was significantly higher than the expression in adjacent normal tissues (1.058±0.636),t =4.001,P<0.05.The relative expression of MIR155HG in LSCC tissues (3.865±1.012)was significantly higher than the expression in adjacent normal tissues (1.686±0.456), t=2.818,P<0.05.The expression of miR-155-5p and MIR155HG in LSCC tissues was significantly correlated with TNM stage,pathological differentiation and lymph node metastasis (P<0.05),but not associated with age,smoke and drink(P>0.05).The expression of MIR155HG was positively correlated with the expression of miR-155-5p in LSCC tissues,r=0.654 3,P<0.05.CONCLUSIONS The genesis,invasion and metastasis of LSCC may be closely related to the increased expression of miR-155-5p and its host gene lncRNA MIR155HG.The expression of miR-155-5p is consistent with the expression of MIR155HG.They may become new tumor marker in targeted therapy of LSCC.
