摘　　要：目的　观察过氧化氢处理对体外培养肠上皮细胞株SW 480线粒体编码基因mRNA的影响。方法　以 40 0 μmol/L和 4mmol/L的过氧化氢处理人肠上皮细胞细胞株 (SW 480 ) 3h后 ,分离RNA ,应用RT PCR检测ATPase6、ATPase8、COI、COII及COIIImRNA的变化。结果　过氧化氢对肠上皮细胞线粒体编码基因mRNA的表达具有明显影响 ,但是这种改变不与过氧化氢的剂量呈现出一种一致的变化。Objective To study the effects of hydrogen peroxide on human mitochondrial gene expression of intestinal epithelial cells by using in vitro model of hydrogen peroxide stimulated SW 480 cell line.Methods Human intestinal epithelial cell were treated with 400 μmol/L and 400 mmol/L hydrogen peroxide and RNA of hydrogen peroxide induced SW 480 cell line was isolated. Changes in ATPase 6, ATPase 8, COI,COII and COIII gene expressions were studied through reverse transcriptional polymerase chain reaction(RT PCR).Results Hydrogen peroxide induced significant changes in mitochondrial gene expression,but the changes were not in consistence with changes in dosage of hydrogen peroxide.Conclusion Differential expressions of mithochondrial gene may be one of the mechanisms involved in hydrogen peroxide induced apoptosis of human intestinal epithelial cells.
