摘  要:为了建立一种适用于治疗研究的急性出血坏死型胰腺炎大鼠模型 ,将 3 %牛磺胆酸钠 1 ml在大鼠胰被膜下均匀注射。结果表明手术后半小时大鼠胰腺出现出血和坏死 ,血清淀粉酶和胰腺组织 TNFα水平在 2 4h内达到高峰 ,并持续至 48h,同时继发肝脏和肺组织的病理损害和 TNFα水平升高。该模型 2 d死亡率为 0 ,以后逐渐升高 ,一周死亡率达90 %。结果提示采用胰被膜下均匀注射 3 %牛磺胆酸钠法 1 ml可建立急性出血坏死型胰腺炎合并多器官损害大鼠模型 ,且胰腺炎病情逐渐加重 ,一周的死亡率稳定 ,与人类重症急性胰腺炎的自然病程相类似 ,或可成为治疗研究的理想的动物模型。To establish a model of acute hemorrhagic-necrotizing pancreatitis (AHNP) in rats for evaluating therapy, the rats were chalanged by homogeneous injection of 1 ml of 3 percent sodium taurocholate beneath the pancreatic capsule. The results showed that pancreatic hemorrhage and necrosis were observed within half an hour of the operation. Serum amylase and pancreatic tissue TNFα levels were peaked 24 hours later and maintained for 24 hours. Histological injury and progressively increased TNFα levels of liver and lungs were observed subsequently. Two-day mortality of this model was zero. The mortality was gradually increased and reached to 90% after one week. The results suggested that the AHNP rat models established were accompanied by multiply organ injury. The severity of AHNP is progressive with finite one-week mortality rate. The model resembles the natural course of human severe acute pancreatitis(SAP) and may be suitable for evaluating therapeutic study on SAP.
