恶性肿瘤自杀基因治疗中GCV和ACV体外抑瘤作用的比较研究

摘　　要：目的 :用含有HSV -TK基因的重组腺病毒/丙氧鸟苷系统(AdTK/GCV)及该重组病毒/无环鸟苷系统(AdTK/ACV) ,在体外分别对4种肿瘤细胞进行抑瘤实验 ,比较GCV、ACV对肿瘤细胞的抑制效果。方法:将同滴度AdTK感染的4种肿瘤细胞分别加入GCV、ACV ,用MTT法测定OD值 ,计算细胞抑制率 ,用SPSS统计软件进行数据分析。结果:相同浓度的GCV与ACV相比 ,对肿瘤细胞的抑制率无显著差异。结论:由于ACV最大耐受量是GCV的4倍 ,且价格低廉 ,故在AdTK自杀基因治疗中可用ACV取代GCV。Objective:To compare the tumor-inhibition effect of ACV and GCV invitro in suicide gene therapy experiment for malignant tumor.Methods:Add ACV and GCV to four tumor cell lines that have been infected with the same TCID50 concentration of recombinant adenovirus containing TK gene(AdTK) individually, then measure OD value by MTT method, calculate the cell-inhibition ratio.Data analysis was done using spss ststistics software.Results:As we observed, there is no significant difference in tumor-inhibition ratio between ACV and GCV on the same concentration.Conclusion:Since the most endurance amount of ACV is four-fold times of GCV and ACV is much cheaper than GCV, so there could be a replacement of GCV by ACV intumor suicide gene therapy.
