骨肉瘤特异杀伤性T淋巴细胞的体外扩增

摘　　要：目的　研究骨肉瘤特异性细胞毒性 T淋巴细胞(CTL)体外扩增培养的方法 ,并进行细胞毒检测 .方法　抽取骨肉瘤患者自身的外周血 ,分离淋巴细胞 ,用 IL- 2体外短期刺激方法提高对骨肉瘤细胞的免疫性 ,再按一定比例体外与多西紫杉醇处理的骨肉瘤细胞及巨噬细胞混合培养(MTL C)可获得骨肉瘤的特异性 CTL 细胞 ,进行鉴定并观察其杀伤特性 .结果　混合培养 4 d后 ,即可扩增出大量淋巴细胞 ,流式细胞仪证实主要是 CD8+ 细胞 ,这些具有杀伤活性的CTL 细胞体外对自身肿瘤细胞有很强的细胞毒作用 .结论　在 IL - 2作用下 ,应用巨噬细胞、自体肿瘤细胞及淋巴细胞体外混合培养 ,是体外扩增特异性细胞毒性 T淋巴细胞的较好方法 ,CTLAIM To induce the osteosarcoma specific CTL proliferation in vitro . METHODS We isolated T lymphocytes from peripheral blood of osteosarcoma cases, which were previously dealt with docitaxol, and activated with combination of macrophages and interleukin 2 (IL 2), then their tumor killing ability was measured. RESULTS A large number of lymphocytes that proliferated after 3 d mixed cul ture were proved mainly to be CD8 + T cell by FCM, and their tumor killing ability was great in vitro . CONCLUSION The specific CTL proliferation in vitro by mixed culture of macrophages, host tumor cells and lymphocytes under the presence of IL 2 is a prospective way in the immunological therapy of tumor .It may improve the therapeutic effect of osteosarcoma.
