异丙酚对大鼠全脑缺血再灌注后海马Akt—FOXO3a信号通路的影响

摘  要:目的探讨异丙酚对大鼠全脑缺血再灌注后海马Akt—FOXO3a信号通路的影响。方法采用四血管闭塞法建立全脑缺血模型(IR组),全脑缺血再灌注+异丙酚组(IR+P组)脑缺血前10min腹腔注射异丙酚,假手术组(S组)仅接受假手术。术后48h三组大鼠进行神经功能学检测。术后4h处死取大脑,SP法进行免疫组化以及Western blots检测大鼠海马Akt、P—Akt、FOX03a、P—FOXO3a蛋白的表达。结果与S组比较,IR组和IR+P组运动功能和感觉功能均明显降低(P〈0.01),IR+P组高于IR组(P〈0.05或P〈0.01)。免疫组化及Western blots检测结果显示,三组海马Akt和FOX03a蛋白表达量在术后比较差异无统计学意义(P〉0.05);IR组和IR+P组海马P—Akt蛋白表达量明显高于s组(P〈0.05或P〈0.01);IR+P组低于IR组(P〈0.05或P〈0.01)。IR组和IR+P组海马P—FOXO3a蛋白表达量明显低于S组(P〈0.01),IR+P组高于IR组(P〈0.05或P〈0.01)。结论异丙酚可能通过对全脑缺血再灌注大鼠海马区Akt—FOXO3a信号通路活化的抑制作用,起到保护大鼠神经功能的作用。Objective To investigate the effects of propofol on hippocampus Akt - FOXO3a signaling pathway of global cerebral ischemia and reperfusion rats. Methods Rat global cerebral ischemic model was established by four - vessel occlusion (IR group ), IR + P group rats got intraperitoneal injection of propofol 10 min before ischemia, sham operation group (S group) received only sham surgery. Neurological functions of three groups were detected 48 h after surgery. Rats were sacrificed and rat brains were obtained 4 h after surgery to detect protein expression of Akt, p - Akt, FOXO3a, p - FOXO3a in rat hippocampus by immunohistochemistry (SP method) and Western blotting. Results Sports functionality and sensory function of IR group and IR + P group were significantly lower than S group ( P 〈 0. 01 ), and were higher in IR + P group than those in IR group (P 〈 0. 05 or P 〈0. 01 ). Immunohistochemistry and Western blots results showed that the expression of Akt and FOXO3a in the three groups showed no significant difference (P 〉 0.05) ; the expression of p - Akt in IR group and IR + P group were significantly higher than that in S group ( P 〈 0.05 or P 〈 0. 01 ), and it was lower in IR + P group than that in IR group (P 〈0.05 or P 〈0.01 ). The expression of p - FOXO3a of IR group and IR + P was significantly lower than that in S group (P 〈0.01 ), and was higher in IR + P group than that in IR group (P 〈 0.05 or P 〈 0. 01 ). Conclusion Propofol possibly affects on hippocampus Akt - FOXO3a signaling pathway of global cerebral ischemia and reperfusion in rats and it plays a protective role in neurological function of rats.
